Evaluation of 2,3,5-triphenyltetrazolium chloride as a stain for detection and quantification of experimental cerebral infarction in rats.

Abstract: SUMMARY We have evaluated the use of 2, 3, 5-triphenyltetrazolium chloride (TTC) as an histopathologic stain for identification of infarcted rat brain tissue. The middle cerebral artery (MCA) of 35 normal adult rats was occluded surgically. At various times after surgical occlusion, rats were sacrificed and brain slices were obtained and stained with TTC or hematoxolin and eosin (H & E); the size of the area of infarcted tissue stained by each method was quantified. In rats sacrificed 24 hr after occlusion of … Show more

“…Degenerating neurons have been shown as early as 30 minutes after a 60-minute MCAO in rats (Chen et al, 2009). When focusing on early reliable detection of the infarct, in particular volumetry, macroscopic inspection using the 2,3,5-triphenyltetrazolium hydrochloride (TTC) stain is a well-established methodological option (Bederson et al, 1986). Strictly speaking, TTC is a marker of functioning mitochondria, which reduce TTC to a red-colored, lipid-soluble formazan.…”

“…TTC interacts with the electron transport chain in the inner mitochondrial membrane of viable tissue and is thereby converted to a red dye (Bederson et al 1986a). The size of the ischaemic infarct volume was calculated using the software program Brain Damage Calculator 1.1 (MB Teknikkonsult, Lund, Sweden).…”

O tratamento atual da isquemia cerebral muitas vezes não é eficaz, devido a este fato vários pesquisadores têm procurado maior esclarecimento de sua fisiopatologia com o objetivo de encontrar um tratamento eficaz 1, 2 .…”
