Evaluation of a Novel Rapid Test System for the Detection of Allergic Sensitization to Timothy Grass Pollen against Established Laboratory Methods and Skin Prick Test

Abstract: Type I hypersensitivity is driven by allergen specific immunoglobulin E (sIgE) and thus sIgE represents a marker for modern allergy diagnosis. Recently, a rapid assay for the detection of sIgE, termed as (Allergy Lateral Flow Assay) ALFA, has been developed. The objective of our study is the evaluation of a scanner-based system for the semiquantitative interpretation of ALFA results. Agreement to Skin Prick Test (SPT, Allergopharma), ALLERG-O-LIQ System (Dr. Fooke), and ImmunoCAP (Phadia) was investigated usin… Show more

“…The sIgE detection method we used was ALLERG-O-LIQ. According to the studies by various authors, the consistency of ALLERG-O-LIQ with ImmunoCAP results (the most common method used in allergy diagnosis) for quantitative or semi-quantitative determination of various allergens has been verified [ 22 - 24 ], although the correlation is stronger for aeroallergens than for food allergens.…”

“…We used class 2 as the cut-off level of positive results for both ImmunoCAP and AdvanSure, as suggested by the manufacturers, in calculating the agreement percentage for positive and negative calls. The cut-off for a positive result from the GOLD chip was determined as the fluorescence intensity corresponding to 0.7 IU/mL of IgE for each allergen extract [ 26 27 28 ]. The overall total agreement rates were 92.2% for GOLD vs Immuno-CAP, 66.1% for ImmunoCAP vs AdvanSure, and 66.1% for GOLD vs AdvanSure ( Table 3 ).…”

“…LFIA is the combination of a labeled immunoassay with chromatography, in which capillary forces force the analyte to move. Specific recognition elements, representing specific binding moieties, are immobilized on the membrane surface and are thus able to detect different analytes, such as allergens [9,10]. LFIAs have several key features [11]: (1) reaction speed is fast, occurring in as little as a few minutes; (2) the assay can automatically separate target analytes from biological matrices without complex additional steps; and (3) the assay can be adapted to suit a variety of cluttered outdoor environments, without the need for highly skilled personnel to operate equipment or carry out complex analytical procedures.…”
