Evaluation of CTB-sLip for Targeting Lung Metastasis of Colorectal Cancer

Abstract: Lung metastasis of colorectal cancer is common in the clinic; however, precise targeting for the diagnosis and therapy purposes of those lung metastases remains challenging. Herein, cholera toxin subunit b (CTB) protein was chemically conjugated on the surface of PEGylated liposomes (CTB-sLip). Both human-derived colorectal cancer cell lines, HCT116 and HT-29, demonstrated high binding affinity and cellular uptake with CTB-sLip. In vivo, CTB-sLip exhibited elevated targeting capability to the lung metastasis o… Show more

“…FAM-loaded sLip (sLip/FAM) and FAM-loaded IR783-sLip (IR783-sLip/FAM) were prepared based on the same approach. Specifically, FAM was dissolved in sterile saline (65 °C, pH 7.0) at first, followed by the same procedure as outlined above, with the final removal of free FAM using a Sephadex G50 column [ 38 ]. The FAM concentration was measured under conditions of excitation (Ex) at 494 nm and emission (Em) at 580 nm using a Spark Cyto automatic microplate reader (Tecan Group Ltd., Männedorf, Zurich, Switzerland).…”

“…Human-derived colorectal cancer cell lines demonstrated high binding affinity and cellular uptake with CTB-sLip. Moreover, these nanosystems showed elevated targeting capability for the lung metastasis of colorectal cancer in a model of nude mice in comparison to pegylated liposomes without CtxB modification [ 147 ]. CtxB applications are summarized in Table 4 .…”

“…Zhang and his team developed PEGylated liposomes (CTB-sLip) coupled with cholera toxin subunit b protein. These CTB-sLips showed excellent affinity and cellular uptake in both human-derived CRC cell lines, including HCT-116 and HT-29 cells [ 81 ]. In another study, immunoliposomes, coated with antibodies to Frizzled 10 protein, were synthesized and loaded with 5-FU.…”
