Evaluation of Detect-A-Strep and the Culturette Ten-Minute Strep ID kits for detection of group A streptococcal antigen in oropharyngeal swabs from children

Abstract: The Detect-A-Strep kit (DAS) (Antibodies Inc.) and the Culturette Brand 10-Minute Group A Strep ID kit (CBTMSI) (Marion Scientific) were compared with a sensitive culture method for detection of group A streptococci in oropharyngeal swabs from children. Specimens from 953 children ranging in age from 4 months to 18 years were inoculated to selective blood agar plates containing sulfamethoxazole-trimethoprim. Culture plates were incubated anaerobically for the first overnight period and aerobically in the prese… Show more

“…Other variables, some of which can be controlled more effectively than others, include the efficiency of swabbing the throat, particularly with children (3), the use of transport medium with the swab, and time delays between sampling and testing (8). Probably of greatest significance is the efficiency of the reference culture procedure in recovering organisms from the samples (1,4). Therefore, results of the different studies should be interpreted with appropriate caution, and the results of this study may not be comparable in absolute terms with those in other publications.…”

“…The original Lancefield precipitation method for the serogrouping of beta-hemolytic streptococci has been largely superseded by more rapid and sensitive methods, such as latex agglutination. Latex agglutination has also been used for the rapid detection of bacteria or antigens in blood cultures (12)(13)(14), body fluids (1,5), and swabs taken from various sites of infection (2,3,7,10). Although previous studies indicate that reasonably reliable results can be obtained, the potential for antigenic cross-reactions among different bacteria has not been fully appreciated.…”

“…A recent study by Carlson et al (4) described ssA as being a superior tool for GABHS isolation. Overnight anaerobic incubation followed by a second overnight incubation period in an enriched CO2 atmosphere was also described (3).…”
