Evaluation of genetic diversity amongst Descurainia sophia L. genotypes by inter-simple sequence repeat (ISSR) marker

Abstract: Descurainia sophia is a valuable medicinal plant in family of Brassicaceae. To determine the range of diversity amongst D. sophia in Iran, 32 naturally distributed plants belonging to six natural populations of the Iranian plateau were investigated by inter-simple sequence repeat (ISSR) markers. The average percentage of polymorphism produced by 12 ISSR primers was 86 %. The PIC values for primers ranged from 0.22 to 0.40 and Rp values ranged between 6.5 and 19.9. The relative genetic diversity of the populati… Show more

“…In recent years, inter-simple sequence repeat (ISSR) have also been utilized as a tool in the study of conservation genetics and outcrossing rates (Han et al, 2009;Saki et al, 2016). ISSR is a technique that uses repeat-anchored primers to amplify DNA sequences between two-inverted SSR (Zietkiewicz et al, 1994).…”

“…Outcrossing rates in plant populations have often performed using codominant markers such as SSR, allozyme (Chen et al, 2009;Suwarni et al, 2018). However, in the absence of sufficient allozyme polymorphism, several dominant markers such as RAPD (Gaiotto et al, 1997;Dong et al, 2006), ISSR (Han et al, 2009;Fernando et al, 2015;Saki et al, 2016), and AFLP (Gaiotto et al, 1997;Pometti et al, 2013., Zhao et al, 2014Dong et al, 2018a;2018b)…”

“…PIC value ranged from 0.38 to 0.46, and the mean value was 0.424. The PIC value ranged from 0.5 to 0.25, considered an informative marker ( Saki et al, 2016 ). Thus all markers in our study are informative markers.…”

“…Inferences about plant mating systems increasingly use highly informative genetic markers and investigate finer facets of the mating system (Ritland, 2002). In recent studies, although several dominant markers, such as RAPD (Gaiotto et al, 1997;Dong et al, 2006), ISSR (Han et al, 2009;Fernando et al, 2015;Saki et al, 2016), and AFLP (Gaiotto et al, 1997;Pometti et al, 2013;Zhao et al, 2014;Dong et al, 2018 b) markers, have been used to estimate outcrossing rates in plant species, outcrossing rates in plant populations have often been determined using codominant markers, such as isozymes and microsatellites, due to their high allelic polymorphism (Chaix et al, 2003;Chen et al, 2009;Adugna et al, 2013;Sinha et al, 2015;Sharma et al, 2017;Suwarni et al, 2018;Zhang et al, 2019;Dauphin et al, 2020;Sun et al, 2020;Torres et al, 2020;Calevo et al, 2021;Heike and Dagmar, 2022;Lu et al, 2022).…”
