Evaluation of Hypoglycemic and Antioxidative Effects of Synthesized Peptide MC62

Abstract: Diabetes mellitus has been considered as a major health problem in the world today. This study aimed to investigate the hypoglycemic and antioxidative effects of peptide MC62 which was synthesized by solid-phase peptide synthesis method against diabetes induced by streptozotocin. MC62 was administered daily and injected intraperitoneally to the diabetic mice at a dose of 1 μmol/kg body weight for 20 days. The levels of fasting blood glucose and HbA1C, pancreatic islet damage, and associated changes in antioxid… Show more

“…Consistent with previous reports, 26,31,32 several parameters of global and pancreatic oxidative stress were changed in STZinduced DM mice. Several parameters indicative of oxidative stress in serum (malondialdehyde (MDA), glutathione (GSH), glutathione peroxidase (GSH-PX), and superoxide dismutase (SOD)) may be altered in STZ-induced DM mice.…”

“…The general procedure of peptide synthesis under microwave irradiation has been reported. [30][31][32] The purity of the crude peptide was 85%. HPLC analyses were undertaken on a 2010-20AT system (Shimadzu, Kyoto, Japan) on a C18 reversed-phase column (5 μm, 250 mm × 4.6 mm) by gradient elution using (A) water with 0.1% TFA-(B) acetonitrile with 0.1% TFA as the mobile phase from 0 min (10% B) to 15 min (90% B).…”

“…Male KM mice , 6–8 weeks old, weighing 18–22 g, were purchased from Comparative Medicine Centre of Yangzhou University, Jiangsu, China, and housed six per cage under the conditions of ambient temperature (24 ± 1 °C) under a reverse 12‐h light/dark cycle, with free access to food and water. The mice were fasted for overnight before the injection of STZ intraperitoneally (40 mg/kg/day) for 5 consecutive days.…”

“…Expression of this gene in tandem repetitions improved the yield of the recombinant MC6.1, which had not been satisfactory in a previous study carried out by Liu et al [111]. In addition to this, the hypoglycemic peptide MC6.2, derived from MC6.1 and composed of 11 amino acid residues, was chemically produced by a microwave peptide synthesizer and used in an assay in which it was intraperitoneally administered daily for 20 days to streptozotocin-induced diabetic mice [113]. This daily administration resulted in lower blood glucose levels measured every day after the treatment in comparison with the control group and also improved some symptoms related to hyperglycemia.…”
