Evaluation of in vitro Probiotic Potential of Pediococcus pentosaceus OZF Isolated from Human Breast Milk

Abstract: This study was conducted to evaluate the probiotic properties of Pediococcus pentosaceus OZF isolated from human breast milk. The results obtained so far suggest that the strain is resistant to low pH, bile salt, pepsin and pancreatin, so it could survive while passing through the upper part of the gastrointestinal tract and reveal its potential probiotic action on host organism. The strain was non-pathogenic (γ-hemolytic), produced anti-Listerial bacteriocin, exhibited a strong autoaggregating phenotype (85.7… Show more

“…For the pathogen (Listeria monocytogenes MTCC 657) exclusion assay, the monolayers were first treated with the LAB isolates (5 × 10 10 -10 11 CFU mL −1 ) for 90 min, washed with PBS to remove non-adhered LAB followed by addition of Listeria cells (2 × 10 10 CFU mL −1 ) suspended in serum and antibiotic free medium and incubated for 60 min [20]. The excluded Listeria cells were recovered by washing with the PBS and were plated on Listeria selective agar [28]. The amount of excluded Listeria cells were calculated as the amount of Listeria cells recovered after incubation compared to initial amount added to the cell lines with the following formula, Amount of excluded Listeria cells = log (X) − log (Y) where X is the no.…”

“…Pediococcus pentosaceus and Lactobacillus salivarius are most frequently isolated from the duodenal mucosa of piglets (Zhang et al, 2011;Zhang et al, 2015) and human breast milk (Osmanagaoglu et al, 2010;Kiran et al, 2015), but little information is available about the isolated strains from animals and their bioavailability. Therefore, the objectives of this study were to isolate and identify P. pentosaceus and L. salivarius strains from cecal content and investigate their potential probiotic effects on growth, meat quality and microenvironment in specific pathogen free (SPF) chickens, and ammonia treatment in vitro.…”
