Olive leaf extract is characterized by a high content of phenols and flavonoids (oleuropein, luteolin, and their derivatives). These compounds are defined as secondary metabolites and exert such as anti-inflammatory, antioxidant, and antimicrobial activities. We investigated the in vitro antifungal activity of two olive leaf extracts (named EF1 and EF2) against a Fusarium proliferatum (AACC0215) strain that causes diseases to many economically important plants and synthesizing diverse mycotoxins. In this work, we aimed to identify the most appropriate concentration between the tested two olive leaf extracts to develop a safe, stable and efficient drug delivery system. Qualitative and quantitative analyses of the two olive leaf extracts by (HPLC) were performed. Furthermore, we also evaluated the antifungal effects of the two leaf extracts when encapsulated in chitosan-tripolyphosphate nanoparticles. The major compound in both EF1 and EF2 was oleuropein, with 336 and 603 mg/g, respectively, however, high concentrations of flavonoid were also present. EF1 and EF2 showed a concentration depended effect on F. proliferatum (AACC0215) viability. Our results showed a great efficacy of EF1/nanoparticles at the higher concentration tested (12X) against the target species. In this case, we observed an inhibition rate to both germination and growth of 87.96 and 58.13%, respectively. We suggest that EF1 olive leaf extracts, as free or encapsulated in chitosan-tripolyphosphate nanoparticles, could be used as fungicides to control plant diseases. Finally, future application of these findings may allow to reduce the dosage of fungicides potentially harmful to human health.

Background: In a prior study, Lasiodiplodia theobromae (Pat.) Griffiths and Maubl. and Bipolaris bicolor (Mitra) Shoemaker., were found to suppress the growth of Eleusine indica (L.) Gaertn, but limited information exists on their response to environmental factors and herbicides for integrated E. indica control.Objective: This study aimed to determine the tolerance levels of L. theobromae and B. bicolor to pH, temperature, photoperiod, relative humidity, and herbicides. Methods: The mycelia and conidia of L. theobromae and B. bicolor were exposed to a range of environmental conditions and herbicides in a controlled setting, including different levels of temperature (25, 30, 35 ⁰C), pH (4, 6, 8, 10), photoperiod (24 hours of darkness; alternating 12 hours of blue light followed by 12 hours of darkness; and alternating cycles of 6 hours of blue light and 6 hours of darkness), and relative humidity (75, 85, 95%). Herbicides such as topramezone, diuron, oxyfluorfen, and imazethapyr were applied at their recommended rates.Conidial germination was assessed by counting after an 18-hour incubation using a haemocytometer, while the diameter of mycelium growth was measured after 3 days of incubation, except for the herbicide effects evaluation, which were extended over 7 days. Results: Laboratory assays demonstrated that L. theobromae exhibited higher conidial germination of 85-95% and superior mycelial growth under varied pH, temperature, and photoperiod conditions compared to B. bicolor. Lasiodioplodia theobromae's mycelia remained unaffected by herbicides at full labelled rates, but they inhibited the conidial germination of the fungus. For B. bicolor, the conidia were not affected by imazethapyr and topramezone, but its mycelial growth was reduced by imazethapyr, oxyfluorfen, and diuron. Conclusions: These results indicate that the mycelia of L. theobromae presents a more favorable option for tank mixing with test herbicides, offering potential for the formulation of an integrated control strategy against E. indica.

Phytopathogenic fungi are the predominant causal agents of plant diseases. Available fungicides have substantial disadvantages, such as being insufficiently effective owing to intrinsic tolerance and the spread of antifungal resistance accumulating in plant tissues, posing a global threat to public health. Therefore, finding a new broad-spectrum fungicide is a challenge to protect plants. We studied the potency of a novel antimicrobial agent, M451, a 1,6-diaminohexane derivative, against different phytopathogenic fungi of the Ascomycota, Oomycota, and Basidiomycota phyla. M451 exhibited significant antifungal activity with EC50 values from 34–145 μg/mL. The minimal fungicidal concentration against Fusarium oxysporum ranged from 4 to 512 μg/mL depending on the exposure times of 5 min to 24 h. M451 has the highest activity and significantly lower exposure times compared to different polyene, azole, and phenylpyrrole antifungals. The conidial germination assay revealed that M451 induced 99 and 97.8% inhibition against F. oxysporum within 5 min of exposure to 5,000 and 500 μg/mL, respectively. Germ tube elongation, spore production, and spore germination were also significantly inhibited by M451 at concentrations of ≥50 μg/mL. Based on the broad spectrum of antifungal effects across different plant pathogens, M451 could be a new chemical fungicide for plant disease management.
