The prevalence of human adenoviruses (HAdV) in river waters was investigated in this study. Water samples were collected from 13 rivers in Taiwan, concentrated, and assessed for the presence of HAdVs using nested polymerase chain reaction (PCR). Human AdV positive samples were then subjected to real-time PCR (qPCR) to quantify the viral genomes and further subjected to primer-based genotyping to identify the various serotypes present. For each water sample, several water quality parameters were evaluated, including heterotrophic plate count, total coliform, Escherichia coli, water temperature, pH, conductivity, and dissolved oxygen. Among the 13 rivers examined, four rivers (30.8 %) were found to contain HAdVs. The major genotype was F species HAdV serotype 41. The mean HAdVs concentrations ranged from 6.10 × 10(2) to 8.51 × 10(2) copies/L. No significant differences were observed between the presence of HAdVs, and all of the water quality parameters evaluated (heterotrophic plate count, total coliform, E. coli, water temperature, pH, conductivity, and dissolved oxygen). Given the potential health risks posed by the presence of enteric viruses in environmental waters, further assessment is desirable with respect to possible sources, virus transport, and survival of viruses in the aquatic environment.

Bacteriophage MS2 is widely used as a surrogate to estimate pathogenic virus elimination by membrane filtration processes used in water treatment. Given that this water technology may be conducted with different types of waters, we focused on investigating the effects of ionic strength on MS2 behavior. For this, MS2 was analyzed while suspended in solutions of various ionic strengths, first in a batch experiment and second during membrane ultrafiltration, and quantified using (i) quantitative reverse transcriptase PCR (qRT-PCR), which detects the total number of viral genomes, (ii) qRT-PCR without the RNA extraction step, which reflects only particles with a broken capsid (free RNA), and (iii) the PFU method, which detects only infectious viruses. At the beginning of the batch experiments using solutions containing small amounts of salts, losses of MS2 infectivity (90%) and broken particles (20%) were observed; these proportions did not change during filtration. In contrast, in high-ionic-strength solutions, bacteriophage kept its biological activity under static conditions, but it quickly lost its infectivity during the filtration process. Increasing the ionic strength decreased both the inactivation and the capsid breakup in the feed suspension and increased the loss of infectivity in the filtration retentate, while the numbers of MS2 genomes were identical in both experiments. In conclusion, the effects of ionic strength on MS2 behavior may significantly distort the results of membrane filtration processes, and therefore, the combination of classical and molecular methods used here is useful for an effective validation of the retention efficiency of ultrafiltration membranes.

L’émergence de pathogènes dans l’eau destinée à la consommation humaine représente une préoccupation majeure en matière de santé publique pour les industriels et les pouvoirs publics concernés. Parmi ces pathogènes, certains sont d’origine fécale (Cryptosporidium, Campylobacter ou bien les rotavirus), alors que d’autres vivent dans l’environnent naturel (Legionella, Pseudomonas, Aeromonas ou bien les mycobactéries). Dans l’optique de mettre en place une analyse des risques liés à la présence de ces pathogènes, il est important d’accroître nos connaissances sur l’écologie de ces microorganismes et de développer des outils d’analyse afin de réaliser une meilleure surveillance sanitaire. Par conséquent, l’écologie microbienne du réseau de distribution d’eau potable doit être étudiée en détail, particulièrement vis-à-vis des propriétés physiologiques et la diversité des espèces microbiennes présentes, afin de mieux comprendre les interactions entre les espèces communément rencontrées et celles pathogènes.The emergence of pathogens in water intended for human consumption is a major concern in terms of the public health industry and the public authorities concerned. Among these pathogens, some are of faecal origin (Cryptosporidium, Campylobacter, or rotavirus), while others live in the natural environment (Legionella, Pseudomonas, Aeromonas or mycobacteria). In order to establish a risk analysis related to the presence of these pathogens, it is important to increase our knowledge on the ecology of these microorganisms and to develop analytical tools to achieve better health monitoring.
