Evaluation of Multiplexed Cytokine and Inflammation Marker Measurements: a Methodologic Study

Abstract: Background Chronic inflammation is etiologically-related to several cancers. We evaluated the performance (ability to detect concentrations above the assay’s lower limit of detection, coefficients-of-variation [CVs], and intraclass correlation coefficients [ICCs]) of 116 inflammation, immune, and metabolic markers across two luminex bead-based commercial kits and three specimen types. Methods From 100 cancer-free participants in the Prostate, Lung, Colorectal, and Ovarian Cancer Trial, serum, heparin plasma,… Show more

“…Indeed, both platforms failed to robustly detect the GM-CSF, IL-1β, IL-2 and IL-5 cytokines for which previous investigations had reported significant variations in expression either during MDE treatment or at baseline in comparison to healthy controls (Biancotto et al, 2013;Dahl et al, 2014;Hernandez et al, 2008;Schmidt et al, 2014;Shelton et al, 2015). These types of inconsistency in the results have already been reported (Breen et al, 2011;Chaturvedi et al, 2011;Dabitao et al, 2011;Dupuy et al, 2013;Fu et al, 2010). In particular, the consensus that the detection of IL-1β suffers from poor performances is of importance.…”

“…There is no absolute biological reason that would mandate similar and/or correlated concentration of all analytes in sera and plasma samples from the same individual (Altara et al, 2015;Chaturvedi et al, 2011;Clendenen et al, 2010;Dossus et al, 2009;RosenbergHasson et al, 2014;Wong et al, 2008). We therefore determined on each platform the degree of correlation between serum and plasma measurements.…”

“…Biomarker levels are affected by method of collection, processing, and storage. 71 All blood specimens were obtained and assayed in fully accredited clinical laboratories which followed established, standardized procedures. We were able to control for a large number of important potentially confounding factors.…”
