Evaluation of neuregulin-1's neuroprotection against ischemic injury in rats using diffusion tensor imaging

Abstract: Stroke is a devastating neurovascular disorder that results in damage to neurons and white matter tracts. It has been previously demonstrated that neuregulin-1 (NRG-1) protects neurons from ischemic injury following stroke. Here, diffusion tensor imaging (DTI) was utilized to characterize the effects of NRG-1 treatment on cererbral infarction and integrity of white matter after ischemic insult using a permanent middle celebral artery occlusion (pMCAo) rat model. In the present study, sixteen Sprague-Dawley rat… Show more

“…Compared with controls, i.a. administration of NRG-1 drastically reduced infarct volume after MCAO in mice as we and others previously showed in rats (Guo et al 2006;Li et al 2007;Shyu et al 2004;Wang et al 2015Wang et al , 2018Xu et al 2004Xu et al , 2005Xu et al , 2006. The percentage of total infarct volume to the ipsilateral hemisphere in vehicle-treated animals was 41%.…”

“…Neuregulins have been implicated in normal brain function, as well as in neuroprotection following cerebral ischemia. Previous studies from our laboratory and others demonstrated that neuregulin-1 (NRG-1) reduced ischemia-induced neuronal death in a rat focal stroke model by~90% with a therapeutic window of > 13 h (Guo et al 2006;Li et al 2007;Shyu et al 2004;Wang et al 2015Wang et al , 2018Xu et al 2004Xu et al , 2005Xu et al , 2006. NRG-1 administration also resulted in a significant improvement of neurological function (Iaci et al 2010;Xu et al 2006).…”

“…Work from our laboratory and others demonstrated that NRG-1 reduced ischemia-induced neuronal death and inflammation in rodent focal stroke models by up to 90% ( Shyu et al, 2004 ; Xu et al, 2004 ; Guo et al, 2006 ; Xu et al, 2006 ; Li et al, 2007 , 2008 , 2009 ; Guan et al, 2015 ; Wang et al, 2015 ; Simmons et al, 2016 ; Ji et al, 2017 ; Surles-Zeigler et al, 2018 ; Wang et al, 2018 ; Noll et al, 2019 ; Cui et al, 2023 ). These finding have been demonstrated by seven independent laboratories.…”

“…NRG1 was transduced through the receptor tyrosine-protein kinase erythroblastic oncogene B (ErbB) family to regulate downstream pathways PI3K–AKT and PI3K–extracellular-signal-regulated kinase signaling pathway. It had been demonstrated NRG1 signaling involved in NPCs activation [ 38 ], neuroprotective effects [ 39 , 40 ], neuroinflammatory modulation [ 41 , 42 ], and angiogenesis [ 43 ]. Through Western blot, the expression of NRG1 and AKT, typically activated in the EBP-bFGF/ECM group, was verified compared with the other 2 groups.…”
