Sea cucumbers are amongst the highest value seafoods available commercially, especially in the south-east Asian region, primarily due to their nutritional and health benefits as applied in Traditional Chinese Medicine. While the majority of studies for nutritional products derived from sea cucumber compounds have been conducted in vitro, the number of in vivo and evidence-based human clinical studies are limited. This review has critically assessed the advances in in vivo and clinical studies of sea cucumber-derived bioactives (both extracts and compounds) via a comprehensive literature research on papers published in the last ten years from 2012-2021. Sea cucumber-derived compounds were reported to have the following activities: anticancer, antihyperlipidemic, antihyperglycemic, anti-inflammatory, anticoagulant/antithrombotic, antioxidant, and antihypertension, immunomodulatory, wound healing and as treatments for Alzheimer’s and Parkinson’s diseases. These active compounds include triterpene glycosides (saponins), fucosylated chondroitin sulfate (FCS), cerebrosides, glycosaminoglycan, fucoidan, phospholipids, polysaccharides, peptides, long-chain bases, Frondanol A5, acid mucopolysaccharide, and phosphatidylcholines. Gaps, challenges and future directions have been identified and discussed separately to progress different areas of research and to further scientific validation, development and application of sea cucumbers for human health and nutritional products.

Memory impairment is becoming a potential health issue with the delicacy of diet and social stress. Sea cucumber peptides (SCP) prevent memory impairment, as previously reported. In this study, further research was performed using hippocampal lysine-acetylome to explore molecular regulation mechanisms. C57BL/6 mice were treated with scopolamine via intraperitoneal injection to simulate memory impairment. To determine the influence of SCP on the total acetylated-protein level of the hippocampus, acetylated-proteomics was performed. SCP increased the acetylation level of histone (H3 and H4). Meanwhile, for non-histones, the differentially acetylated proteins were involved in multiple memory-related pathways, as shown by KEGG enrichment analysis. Additionally, long-term potentiation was confirmed by western blotting. Finally, a combined analysis of proteome and lysine acetylome revealed that SCP contributed to synaptic vesicle cycle regulation and dopamine metabolism. Consequently, our findings revealed that SCP was potentially neuroprotective by regulating post-transcriptional hippocampal protein acetylation.

Sea cucumber is a precious and nutritious food with abundant protein and other bioactive compounds, among which sea cucumber peptide (SCP) exhibits many beneficial functions. Although research on the development of SCP is attracting more attention, the ability to translate these new findings into industrial practice and new products remains immature. This review summarizes the production and characterization of SCP and emphasizes recent advances in its bioactive properties, including antioxidative, angiotensin‐I converting enzyme‐inhibitory, immunomodulatory, anticancer, antifatigue, and neuroprotective effects. Meanwhile, the challenges and prospects of SCP development have been discussed in both scientific research and commercialization. SCP shows great potential in health promotion, yet several challenges need to be solved: the validation of health claims, clarification of its mechanisms, innovations to boost its bioavailability, and improvement of industrial production and sensory properties of end‐products. Hopefully, this information will provide a comprehensive perspective on developing SCP nutraceuticals and functional foods.
