Evaluation of the Antianginal Effect of Isosorbide-5-Mononitrate in Patients with Chronic Stable Angina Pectoris

Abstract: The antianginal and anti-ischemic properties of isosorbide-5-mononitrate (ISMN) were evaluated in 40 patients with chronic stable angina pectoris in a randomized double-blind parallel-group placebo-controlled study. After 2 weeks’ placebo run-in period the patients were randomized to either ISMN, orally 20 mg 2–3 times daily (titrated) or placebo. They underwent bicycle exercise stress test at the end of the placebo period and after 4 weeks of treatment. Compared with placebo ISMN increased the exercise perfor… Show more

“…Several clinical studies report that chronic ISMN treatment does not result in drug tolerance. 6,7,18) These studies suggest that ISMN is an effective antianginal agent for patients chronically treated with nitrates. However, there are few papers that have confirmed experimentally whether GTN is effective in relieving an anginal attack in patients chronically treated with ISMN.…”

“…4,5) ISMN is clinically used for treatment of patients with hypertension or angina pectoris. [6][7][8] Sublingual dosing of nitroglycerin (glyceryl trinitrate; GTN) is used for relief of the anginal pain of patients treated chronically with ISMN. Many clinical reports show lack of development of tolerance to ISMN.…”
