Evaluation of the Effect of Pentoxifylline on Cisplatin-Induced Testicular Toxicity in Rats

Abstract: Chemotherapy is associated with male infertility. Cisplatin (cis-diamminedichloro-platinum (II) (CDDP) as a chemotherapy medication used to treat a number of cancers has been reported to most likely induce testicular toxicity. Administration of antioxidants, such as pentoxifylline (PTX) may reduce some Adverse Drug Reactions (ADRs) of CDDP. Therefore, this study investigated the potentially protective effects of PTX on CDDP-induced testicular toxicity in adult male rats. For this purpose, 42 male rats were ran… Show more

“…Cisplatin significantly increased testis O2°- (Fig 2A), MDA ( Fig 2B) and carbonylated proteins ( Fig 2C). Cisplatin has been described to induce testicular toxicity with stimulation of the production of free radical [15]. This has been confirmed in our study where we have detected in cisplatin-treated rats increased production of anion superoxide in the testicles.…”

“…It is important to use agents having various biological effects such as antioxidant properties in various organ toxicity models experimentally formed with chemotherapeutic drugs. Until today in numerous researches, it used various agents to reduce or mitigate cisplatin‐induced testis toxicity (Fallahzadeh et al, ; Reddy et al, ). This study was presented to explain the potential ameliorative role of eugenol in reducing of cisplatin‐induced testis toxicity on count and morphology of spermatozoa in rats.…”

“…Several studies have implicated increased generation of reactive oxygen and nitrogen species and the consequent impairment in antioxidant defense systems in drug-induced toxicity. 32 Oxidative stress and lipid peroxidation has emerged to play a critical role in the pathogenesis of MTX-induced toxicity. Interestingly, the single administration of MTX (20 mg/kg) to rats induced prominent alterations in antioxidant defense mechanisms.…”
