Evaluation of the Effectiveness of a Chronic Ocular Hypertension Mouse Model Induced by Intracameral Injection of Cross-Linking Hydrogel

Abstract: Background: Glaucoma is an irreversible and blinding neurodegenerative disease that is characterized by progressive loss of retinal ganglion cells. The current animal models of glaucoma fail to provide a chronic elevated intraocular pressure and cannot maintain the optical media clarity for a long time, which brings some difficulties to the study of glaucoma. Here, we developed a new chronic ocular hypertension model of mice induced by cross-linking hydrogel intracameral injection. Methods: C57BL/6J mice aged 6… Show more

“…COH was induced in the right eyes of mice based on the protocol described in our previous study 18 . Briefly, the mice were anesthetized by the intraperitoneal administration of 16 mg/kg xylazine and 80 mg/kg ketamine hydrochloride (Sigma‐Aldrich, St. Louis, MO, USA).…”

“…No obvious retinal change was observed in the injected eye of the PBS group and in the non-treated eyes of all three groups. retina after hydrogel-induced chronic IOP elevation (46), whereas scutellarin may inhibit the inflammatory processes of retinal neurodegeneration through the NLRP3 inflammasome signaling pathway, including NLRP3, apoptosis-associated speck-like protein containing a caspase recruitment domain, cleaved caspase-1, IL-1β, and IL-18 (21), it is possible that the mechanism of the reduced retinal thinning in our scutellarintreated, hydrogel-injected eyes involved intervention of neuroinflammation via the NLRP3 inflammasome pathways. Scutellarin was also found to inhibit abnormally activated microglia and provide protection against neurodegeneration in the eye and the brain (21,47,48).…”

“…Among them, rodent glaucoma models are frequently used due to the ease and low cost of implementation. Most currently available rodent models are accomplished by laser treatment of the outflow area, 13 15 cautery or affliction of osmotic damage to the episcleral and vortex veins, 16 , 17 external ocular compression using circumlimbal suture, 18 20 or injection of beads 21 28 or hydrogel 33 38 into the anterior chamber.…”
