Evaluation of the Lyra Direct Strep Assay To Detect Group A Streptococcus and Group C and G Beta-Hemolytic Streptococcus from Pharyngeal Specimens

Abstract: bThe Lyra Direct strep assay was compared to culture for its ability to detect Streptococcus group A and ␤-hemolytic groups C/G using rapid antigen-negative pharyngeal specimens (n ‫؍‬ 161). The Lyra assay correctly detected all ␤-hemolytic streptococci (group A, n ‫؍‬ 19; group C/G, n ‫؍‬ 5). In batch mode, the Lyra assay reduced intralaboratory turnaround time by 60% (18.1 h versus 45.0 h) but increased hands-on time by 96% (3 min 16 s versus 1 min 40 s per specimen). Various species of Streptococcus cause a… Show more

“…After 7 days, any residual specimens were stored at Ϫ70°C for PCR testing. Additional PCR testing for GAS and group C/G streptococci (Lyra Direct Strep kit; Quidel Corporation, San Diego, CA) was performed for the study per the manufacturer's recommendation (28).…”

“…The illumigene group A Streptococcus (Meridian Bioscience, Inc.) utilizes loop-mediated isothermal amplification (LAMP) technology and has reported sensitivity and specificity of 82% to 99% and 93% to 99.6%, respectively (20,21). The Quidel Corporation has the Solana Strep Complete assay for group A and group C/G detection and the Solana GAS assay (sensitivity, 91 to 98%; specificity, 97 to 98% [22,23]) that utilize helicasedependent amplification for GAS detection, and the Lyra Direct Strep assay, which is a multiplex real-time PCR assay for detection of group A and C or G Streptococcus (sensitivity and specificity, 100%) (24). The Alere i Strep A and Alere i Strep A2 assays (Abbott Laboratories) use the isothermal nicking enzyme amplification reaction (NEAR) and have reported sensitivity and specificity of 96% to 100% and 91.3% to 100%, respectively (25)(26)(27).…”
