Evaluation of the representative of using rumen fluid samples from lambs fed pelleted TMR for analysis of prokaryotic communities

Abstract: The pelleted TMR pulverized the grass during processing, which may result in more solid attached microorganisms in the filtered rumen fluid. The objective of this study was evaluating the necessity of distinguishing physical phases of rumen contents for analysis of prokaryotes communities in rumen of lambs fed pelleted TMR, considering the dissimilarity of diversity and community of bacteria and archaea between fluid and mixed rumen contents. The yield of microbial DNA, bacterial diversity, abundances of fibro… Show more

“…Ruminal epithelial bacteria represent specific microbial ecological niches different from those associated with rumen contents, which function specific roles in recycling of epithelial tissue, scavenging of oxygen, and hydrolysis of urea ( 51 ). Our findings indicate that the inclusion of alfalfa hay increased the relative abundance of Succiniclasticum , Megasphaera, Alloprevotella , Succinivibrio , and Suttonella attached to rumen epithelia ( 52 ). Succiniclasticum ( 53 ) and Succinivibrio ( 40 ) are pivotal bacteria involved in the degradation of fiber and the production of succinic acid.…”

“…In this experiment, it has been shown that the main phyla present in all samples were Firmicutes , Bacteroides , Spirochaetota , Fibrobacterota , and Patescibacteria . Research has indicated that, despite its small genome, the Spirochaetota displays a broad spectrum of potential functional variety, adjusting to the breakdown of intricate polysaccharides within plant cell walls [ 58 ], in addition to synthesizing B-complex vitamins and breaking down proteins in the rumen [ 59 ]. Our investigation revealed that the proportionate abundance of Spirochaetota in the LBL group surpassed that in the CON group.…”
