Evaluation of the Safety and Protection Efficacy of spiC and nmpC or rfaL Deletion Mutants of Salmonella Enteritidis as Live Vaccine Candidates for Poultry Non-Typhoidal Salmonellosis

Abstract: Salmonella enterica serovar Enteritidis (S. Enteritidis) is a host-ranged pathogen that can infect both animals and humans. Poultry and poultry products are the main carriers of S. Enteritidis, which can be transmitted to humans through the food chain. To eradicate the prevalence of S. Enteritidis in poultry farms, it is necessary to develop novel vaccines against the pathogen. In this study, we constructed two vaccine candidates, CZ14-1∆spiC∆nmpC and CZ14-1∆spiC∆rfaL, and evaluated their protective efficacy. … Show more

“…Moreover, the mutant strain was recovered in lower numbers throughout the sampling period, compared to the wild-type strain. The result is in accordance with the previous reports on the deletion of the spiC gene in the S. Pullorum or S. Enteritidis ( Li et al., 2019 ; Wang et al., 2021 ). In addition to being attenuated, the live attenuated Salmonella vaccine should have no side effects in the poultry flocks ( Yin et al., 2015 ).…”

“…The overall eradication of S. Enteritidis in host tissues can prevent horizontal and vertical transmission of bacteria, which is a crucial index for evaluating the protective efficiency of vaccines ( Tang et al., 2019 ). Live attenuated Salmonella vaccines have been confirmed to show a strong ability to eliminate pathogens in the liver, spleen, and ileum but not in the cecum ( Li et al., 2019 ). Reported subunit vaccines such as FliC or SPI-1 effectors also had limitations in clearing Salmonella in the cecum ( Desin et al., 2011 ; Wisner et al., 2011 ; Okamura et al., 2012 ).…”

“…Triple mutant targeting porin, carbohydrate metabolism, and type III secretion system were used by Li et al [288]. The construct combined deletion from SPI-2 (∆spiC) with OmpD porin (∆nmpC) or rfaL involved in mannitol utilization (∆rfaL) in a chicken model of S. Enteritidis infection.…”
