Evidence for a Humoral Mechanism in Volume Expansion Natriuresis

Abstract: A B S T R A C T The role of a humoral mechanism in the natriuresis induced by volume expansion was evaluated using an isolated dog kidney perfused by a second dog which had been pretreated with desoxycorticosterone acetate (DOCA). Expansion of the perfusion dog with an equilibrated volume of blood from a reservoir, resulted in an increase in UNaV (sodium excretion) from 153.6 +27.9 (SEM) to 345.5 ±57.8 AsEq/min, P < 0.001. FENa (fractional sodium excretion) increased from 3.4 +0.6 to 8.1 ±1.2%, P < 0.01. The n… Show more

“…Pieces of bovine brain encompassing the hypothalamic region and excluding the pituitary were dissected from freshly slaughtered cattle (Snider Brothers, Wilkinsonville, MA), frozen immediately on dry ice, and maintained frozen until 9 kg of tissue was obtained. Then 4.5 kg of tissue was thawed, homogenized in, 4 M acetic acid, and extracted with acetone and petroleum ether by the method of Carraway and Leeman (12).…”

“…Physiologic studies provided considerable evidence for existence of such a "third factor" capable of modulating urinary sodium excretion independently of filtered sodium load and mineralocorticoid activity (2)(3)(4). Assays in vitro of plasma and urine extracts from subjects and animals with expanded extracellular fluid volume (5,6), certain chronic uremic patients (7), and animals in mineralocorticoid "escape" (8) confirmed the biological effect of sodium transport inhibition across living cell membranes.…”

“…In a recent report (8) from this laboratory we presented phenomenological evidence in support of the hypothesis that the natriuresis associated with blood volume expansion is mediated in part by a humoral factor. The present investigation was designed to test the hypothesis that suppression of this factor contributes to the antinatriuresis of caval constriction.…”

“…INCE the early work of de Wardener 1 in 1961 on the third factor with a hormonal nature, 2 which controls sodium excretion through inhibition of tubular Na + ,K + -adenosine triphosphatase (ATPase), 3 ' 4 numerous studies have been performed in an attempt to purify and identify chemically such a compound(s). As it is well established that Na + ,K + -ATPase is specifically inhibited by digitalis compounds, it was tempting to speculate that endogenous digitalislike compound (EDLC) or compounds might be relevant and related to the third factor.…”
