Evidence for Detrimental Cross Interactions between Reactive Oxygen and Nitrogen Species in Leber’s Hereditary Optic Neuropathy Cells

Abstract: Here we have collected evidence suggesting that chronic changes in the NO homeostasis and the rise of reactive oxygen species bioavailability can contribute to cell dysfunction in Leber's hereditary optic neuropathy (LHON) patients. We report that peripheral blood mononuclear cells (PBMCs), derived from a female LHON patient with bilateral reduced vision and carrying the pathogenic mutation 11778/ND4, display increased levels of reactive oxygen species (ROS) and reactive nitrogen species (RNS), as revealed by … Show more

“…There is some evidence suggesting that the exposure to high nitric oxide concentrations could impair in vivo the ability to cope with the oxidative stress caused by the genetic defect, thereby driving the pathology in LHON. This was described by Flabella et al in one patient carrying the 11788/ND4 mutation [96]. This same increase in ROS was described in MS as previously discussed [3,50,51].…”

“…Bioavailability of NO at cell level depends on the activity of the NOS isoforms, so that physiological pathways ascribable to eNOS activity are featured by a limited (nM) amount of NO, whereas higher NO concentrations ( μ M) resulting from the activation of iNOS are responsible for structural modifications such as membrane lipid peroxydation and protein nitrosation. These modifications are associated to the onset and maintenance of severe inflammatory states, including atherosclerosis [ 34 , 55 , 56 ].…”

“…Peripheral blood mononuclear cells (PBMC) are used to establish lymphoblast cell lines that have mainly been used for biochemical analyses, viability assays, or to compare the degree of OXPHOS (oxidative phosphorylation) impairment in LHON. PBMCs, similarly to RGCs, have a metabolism mainly sustained by OXPHOS and therefore represent a good model to be investigated in LHON research (Falabella et al 2016).…”
