Evidence of Singlet Oxygen Participation in the Chlorophyll-Sensitized Photooxidation of Indoleacetic Acid

Abstract: Chlorophyll-sensitized photooxidation of indoleacetic acid (IAA)-with chlorophyll extracted from Pisum sativum L. cv. Alaska W.R.-was determined in the presence of deuterium oxide and known quenchers of singlet oxygen ('02) to explore the involvement of '02 in the reaction. 02 uptake was measured in light in a buffered aqueous miceliar system containing Triton X-100, KCI, chlorophyll, and IAA. The rate of 02 uptake was zero in darkness. The reaction was stimulated by deuterium oxide and inhibited by sodium az… Show more

“…Since IAA is widely reported as being susceptible to photo-oxidation, results on lightenhanced conversions should be interpreted with caution. Koch et al (1982) detnonstrated chlorophyll-sensitized photo-oxidation of IAA using micellar preparations containing chlorophyll extracted from pea leaves. In this present study, hoNvever, a negligible proportion of the IAA degraded by chloroplast preparations could be directly attributed to light-induced breakdown of IAA.…”

“…Chlorophyll sensitized photo-oxidation [9], peanut peroxidase [10], horseradish, and tobacco peroxidases [11] catalysed oxidation of IAA have been studied. Electrochemical and peroxidase oxygenmediated oxidation of IAA at physiological pH has been reported [12].…”

“…It fluoresces with a maximum at 590 nm, with a quantum yield of 0.04 and a lifetime of 0.87 ns. At lower pH the main absorption corresponding to the diprotonated form of the triplet state, 3 PSH 2 2+ is located at 660 nm, while it is at 410 nm for the neutral triplet of the dye, 3 PS, observed at pH > 9. 39 Fig .…”
