Excretory/secretory proteins inhibit host immune responses by downregulating the TLR4/NF-κB/MAPKs signaling pathway: A possible mechanism of immune evasion in parasitic nematode Haemonchus contortus

Abstract: Haemonchus contortus is an important parasitic nematode of ruminants. Previous studies showed that H. contortus escape the immunity through complex mechanisms, including releasing excretory/secretory proteins (ESPs) to modulate the host immune response. However, the detailed mechanism through which H. contortus excretory/secretory proteins (HcESPs) promote immune evasion remains unknown. In the present study, we demonstrated that HcESPs inhibit the adaptive immune response of goats including downregulation of … Show more

“…In the near future, controlling some parasites with current anthelminthic medications like oxfendazole, levamisole, and ivermectin may become more challenging ( 10 ). H. contortus has evolved different strategies to evade host immune response during infection ( 12 , 13 ). Parasitic nematodes cause a long-term infection in the host, generally with just a modest inflammatory response.…”

“…The TLR4/NF-κB pathway is a key signaling pathway in the immune system for detecting and responding to infection, inflammation, and external stimuli ( 13 ). This pathway involves multiple molecules and cytokines, including the TLR4 receptor, cascade-activated proteins in the pathway, and ultimately nuclear factor-κB (NF-κB), which is critical for the regulation of inflammation and immune responses ( 14 ). Senescence is a major risk factor for osteoporosis development ( 15 ), which can affect the gut microbial composition, cellular autophagy, and iron metabolism.…”

“…These receptors promote a series of immune responses, including the infiltration of inflammatory cells, the activation of NK cells, and the production of antibodies, thereby enhancing the host's defense capabilities against worm invasion and parasitism [10]. Current research on the interaction between H. contortus and host pattern recognition receptors is still in its infancy, with only a few studies involving CLRs, NLRs, and TLRs [11][12][13].…”
