Exercise Tests and BBB Method for Evaluation of Motor Disorders in Rats after Contusion Spinal Injury

Abstract: Contusion spinal injury of different severity was induced by the weight drop method in male rats by dropping standard weight from the heights of 6.5, 12.5, 25, and 50 mm on the spine after laminectomy at the Th9 level. The dynamics of recovery of voluntary movements was evaluated over 8 weeks after the operation by comparing the traditional semiquantitative BBB score with objective parameters of exercise tests. Exercise tests detected cessation of spontaneous recovery of impaired functions 1-2 weeks earlier th… Show more

“…After skin incision, laminectomy at the Th8 vertebral level was performed. The impact rod (diameter 2 mm, weight 10 g) of an impactor was centered above Th8 and dropped from a height of 25 mm to induce SCI [ 20 ]. Immediately after laminectomy or SCI, in animals receiving injections, UCB-MCs were injected intraspinally into two points (5 μL/injection; 1 × 10 6 cells/5 μL DPBS).…”

“…Weight-drop injury was induced us-ing a standardized instrument that releases a weight (10 g, rod diameter 2 mm) from a height of 2.5 mm onto the exposed dura mater of the spinal cord, which inflicts a moderate contusion injury. 18…”

“…The impact rod (weight 10 g, diameter 2 mm) of the impactor was centered above Th8 and dropped from a height of 25 mm to induce moderate SCI. 22 Following SCI, some of the rats received an intraspinal injection, and then the dorsal back musculature and the skin were sutured. After the surgical procedure, rats received daily doses of gentamicin (25 mg kg − 1 , Russia) intramuscularly for 7 consecutive days.…”
