Existence of novel β‐1,2 linkage‐containing side chain in the mannan of Candida lusitaniae, antigenically related to Candida albicans serotype A

Abstract: The antigenicity of Candida lusitaniae cells was found to be the same as that of Candida albicans serotype A cells, i.e. both cell wall mannans react with factors 1, 4, 5, and 6 sera of Candida Check. However, the structure of the mannan of C. lusitaniae was significantly different from that of C. albicans serotype A, and we found novel b-1,2 linkages among the side-chain oligosaccharides, Manb1fi2Manb1fi 2Mana1fi2Mana1fi2Man (LM5), and Manb1fi2Man-b1fi2Manb1fi2Mana1fi2Mana1fi2Man (LM6). The assignment of thes… Show more

“…Mannans from Serotype A C. albicans also have b-1, 2-linked mannose residues attached to its a-1, 2-linked oligomannose side chains ( Figure S1G; Cutler, 2001;Shibata et al, 2003). These b-1, 2-linked mannose residues are not synthesized when this fungus is cultured in a carbon-limiting low pH medium at a low temperature (Shinohara et al, 2006).…”

“…Glycan was prepared from mannoprotein of the blastospore (yeast) form using Fehling's solution, as previously described (24). The glycans used in this study were purified from strains of C. albicans J-1012 (25), C. albicans NIH B-792 (22), Candida stellatoidea (24), Candida parapsilosis (22), C. lusitaniae (23), S. cerevisiae (WT) (1), S. cerevisiae (mnn1/mnn4) (1), and S. cerevisiae (mnn2) (21). The ␣-mannosidase treatment of C. albicans J-1012 mannan was carried out in 50 mM sodium acetate buffer (pH 4.6) containing 20 units of ␣-mannosidase at 37°C for 48 h.…”

“…C. albicans serotype B mannan acid-stable moiety mannoside units are bound solely through the α-glycosidic bonds. On the contrary, the acid-stable part of C. albicans serotype A mannan contains also β-1,2-linked mannosides at the non reducing ends of α-oligomannoside side chains and the amount of acid-stable β-1,2 linkage containing side chains did not change by yeast to hyphal form transition [6][7][8]. So far the attention in development of anti-Candida vaccine was paid mainly to β-1,2-linked oligomannosides [9,10].…”
