Exopolysaccharide production by mucoid and non-mucoid strains ofBurkholderia cepacia

Abstract: Thirteen strains of Burkholderia cepacia from various origins with mucoid and non-mucoid phenotypes were assayed for exopolysaccharide (EPS) production. The EPS were characterized by glycosyl composition analysis and examination of the products resulting from lithium-ethylenediamine and Smith degradations. The results showed that all strains, including the non-mucoid strains, were able to produce EPS exhibiting the same structural features, i.e. presence of one rhamnosyl, three galactosyl, one mannosyl, one gl… Show more

“…EPS were extracted mostly as described [19]. 30 ml of late log phase cultures (OD 580  = 0.7) grown for three days under standard conditions were harvested by a 30 min centrifugation (5,000 rpm) at room temperature (RT°) to separate the supernatants containing the r eleased EPS ( R PS) from the cell pellets containing the c apsular EPS ( C PS) attached to the cells.…”

“…Expression of capsular polysaccharide by Bcc isolates has been suggested to influence strain virulence and, like the expression of O-PS, has been demonstrated to depend on environmental cues ( Chung et al., 2003 ; Ruskoski and Champlin, 2017 ). Isolates expressing capsule are described in the literature as being “mucoid”, while isolates lacking capsule are described as “non-mucoid” ( Cerantola et al., 2000 ; Ruskoski and Champlin, 2017 ).…”

“…EPS was extracted from the bacterial isolates employing an ethanolic extraction method [21]. Mucoid colonies obtained from 24 h cultures on plates of denitrifying agar medium supplemented with sucrose instead of citrate were scraped with a sterile glass slide and re-suspended in a sterile saline solution (0.85% NaCl).…”
