Exosomal lncRNA NEAT1 from cancer-associated fibroblasts facilitates endometrial cancer progression via miR-26a/b-5p-mediated STAT3/YKL-40 signaling pathway

Abstract: Cancer-associated fibroblasts cells (CAFs) confer a rapid growth and metastasis ability of endometrial cancer (EC) via exosomes-mediated cellular communication. Long non-coding RNA nuclear enriched abundant transcript 1 (lncRNA NEAT1) drives the malignant phenotypes of EC cells. However, the role of exosomal NEAT1 from CAFs in EC progression remains ambiguous, which needs to be investigated. In our study, NEAT1 and YKL-40 were up-regulated, while miR-26a/b-5p was down-regulated in EC tissues. Moreover, NEAT1 e… Show more

“…Previous reports have suggested that CHI3L1 is expressed by and secreted from cancer cells ( 31 - 33 ). Furthermore, CHI3L1 has been reported to serve a role in cancer progression by CAFs; however, this mechanism has only been investigated in the context of immune cells or exosomes, not cytokines ( 11 , 35 ). Accordingly, the present study focused on cytokines secreted from CAFs and demonstrated that CAFs, not cancer cells, mainly secreted CHI3L1 and IL-8.…”

“…Chitinase 3-like 1 (CHI3L1), a 40 kDa secreted glycoprotein also known as YKL-40 ( 25 , 26 ), was originally considered to be associated with inflammatory diseases, such as asthma, liver fibrosis and arthritis ( 27 - 29 ). A number of reports have described the involvement of CHI3L1 in cancer ( 11 , 30 - 35 ). For example, serum CHI3L1 levels are associated with prognosis in patients with colorectal cancer ( 30 ), and upregulation of CHI3L1 expression in tumors promotes tumor angiogenesis, proliferation, migration, invasion and radiation resistance ( 31 - 33 ).…”

“…The downregulation of miR-31 in CAFs has been associated with a corresponding increase in expression of the homeobox gene SATB2 in the context of tumor cell migration and invasiveness in endometrial cancers [62,63]. An exosomal NEAT1 from CAFs has been reported to contribute to endometrial cancer progression via miR-26a/b-5p-mediated STAT3/YKL-40 pathway [64]. A direct transfer of CAF-secreted exosomal miR-320a to endometrial cancer cells has been demonstrated to inhibit their proliferation via downregulation of HIF1alpha, which also led to lowered VEGFA expression in vitro [65].…”

“…Song et al demonstrated that adipocyte-derived exosomes-mediated SNHG9 inhibited inflammation and apoptosis of endothelial cells via downregulating TRADD [ 12 ]. Previous studies indicated that exosomal NEAT1 acted as a vital regulator in the pathogenesis of various diseases, such as autoimmune disorders [ 13 ], malignancies [ 14 ], and cardiovascular diseases [ 15 ]. Nevertheless, the involvement of exosome-mediated transfer of NEAT1 in AR is unclear.…”
