Exosomes isolated from melatonin-stimulated mesenchymal stem cells improve kidney function by regulating inflammation and fibrosis in a chronic kidney disease mouse model

Abstract: Chronic kidney disease (CKD) is defined as structural and functional abnormalities of the kidney due to inflammation and fibrosis. We investigated the therapeutic effects of exosomes secreted by melatonin-stimulated mesenchymal stem cells (Exocue) on the functional recovery of the kidney in a CKD mouse model. Exocue upregulated gene expression of micro RNAs (miRNAs) associated with anti-inflammatory and anti-fibrotic effects. Exocue-treated groups exhibited low tumor necrosis factor-α and transforming growth f… Show more

“…Moroni et al 15 investigated the expression of melatonin receptors in human BMSCs, which are the target cells for melatonin target cells for melatonin, as they have a higher ability to differentiate towards cartilage stem cells than such of other tissues. 16 Yea et al 17 evidenced that TGF-β1 induces the differentiation of BMSCs into chondrocytes to further repair articular cartilage damage. Jin et al confirmed that TGF-β1 was overexpressed in melatonin-treated chondrocytes.…”

“…Accumulating evidence have confirmed the multiple functions of EVs, which have established their potentials as therapeutic mediators for tissue repair and regeneration, 36 including wound healing, 37 bone and cartilage regeneration, 38 , 39 bladder transplantation, 40 kidney, and cardiovascular therapeutics. 41 , 42 As for the application of EVs for dentin regeneration, several studies revealed that exosomes derived from human dental pulp stem cells (DPSCs) and SCAPs could promote dentin-pulp complex regeneration and odontogenic differentiation of DPSCs through TGFβ1/smads and P38/MAPK signaling via transferring microRNAs. 43 45 Zhang and Wang et al 46 , 47 also reported that EVs derived from Hertwig’s epithelial root sheath cells and Schwann cell were able to promote dentin-pulp complex regeneration.…”

“…Melatonin promotes the excretion of exosomes ( 68 ), and it promotes exosome secretion from mesenchymal stem cells also ( 69 ). Early embryonic blastomeres are totipotent stem cells ( 70 ).…”
