Expanded roles of lactate-sensing LldR in transcription regulation of the Escherichia coli K-12 genome: lactate utilisation and acid resistance

Abstract: LldR is a lactate-responsive transcription factor (TF) that transcriptionally regulates the lldPRD operon consisting of lactate permease and lactate dehydrogenase. The lldPRD operon facilitates the utilisation of lactic acid in bacteria. However, the role of LldR in whole genomic transcriptional regulation, and the mechanism involved in adaptation to lactate remains unclear. We used genomic SELEX (gSELEX) to comprehensively analyse the genomic regulatory network of LldR to understand the overall regulatory mec… Show more

“…RNA purity was verified using electrophoresis on a 1.2% agarose gel with formaldehyde, and followed by GelRed-staining. Northern blot analysis was performed as described previously [ 32 ]. Digoxigenin (DIG)-labelled probes were prepared using PCR amplification using BW25113 genomic DNA as a template with a pair of primers ( Table S1b ), DIG-11-dUTP (Roche, Basel, Switzerland), dNTP, gene-specific pair primers, and Ex Taq DNA polymerase.…”

“…factor LldR, a member of the GntR family. LldR is capable of sensing l-lactate [44]. It derepresses the PLldR promoter, and actives the expression of LuxCDE in the presence of Lactate, located on the HCP.…”
