Experimental Research on Differentiation-Inducing Growth of Nerve Lateral Bud by HUC-MSCs Chitosan Composite Conduit

Abstract: This study is intended to explore the role of human umbilical-cord-derived mesenchymal stem cells (HUC-MSCs) in nerve end-to-side anastomosis, as well as in the induction and promotion of growth of nerve lateral bud. The chitosan nerve conduit was prepared based on the biological characteristics of chitosan, and the nerve conduit was filled with HUC-MSCs, and was used to bridge the nerve end-to-side anastomotic stoma. The experimental animals were randomly assigned into three groups (10 in each group), and the… Show more

“…2-The number of medullated fibers and the myelin sheath thickness in group C were higher than those in group A or B. 3-A large number of brown-red proliferating Schwann cells was found in group C but not in others [ 94 ] Dog Sciatic Human umbilical cord mesenchymal stem cells (HUCSCs) 35 mm of the nerve was removed 1 × 10 6 HUCSCs (LOCC) “longitudinally oriented collagen conduit” Electrophysiological measurements, Electron microscopy, Histological analyses & Muscle mass analyses 1-9 months after surgery, nerve conduction was better in the LOCC/HUCSCs group than in the LOCC-alone group. 2-In the middle of the regenerated segment in the LOCC/ HUCSCs group, GAP-43-positive, NF-positive, and S-100-positive cells were found 9 months after surgery.…”

“…The tibial nerve itself then served as a scaffold for the reinnervation of the peroneal innervation field by lateral budding. Results showed that the UC-MSCs played a key role in the induction and growth of the lateral nerve bud, leading to a faster and better recovery, as indicated by significantly better electrophysiological recovery, as well as a high number of myelinated axons [44].…”

“…This disadvantage of a foreign body sensation does not occur if the implant is biodegradable and thus vanishes over time. Examples of such scaffolds are chitosan conduits, which are used either alone [44] or in combination with poly(lactic-co-glycolic acid) (PLGA) fibers [4,43,45,46] or collagen [47]. Another artificial biodegradable conduit used is made of poly L-lactic acid and ε-caprolactone copolymer (PLAC) and is also filled with collagen [48][49][50].…”
