Exploring oxidovanadium(iv) homoleptic complexes with 8-hydroxyquinoline derivatives as prospective antitrypanosomal agents

Abstract: [VIVO(L-H)2] and [VVO(OCH3)(L-H)2] compounds of 8-hydroxyquinoline derivatives L showed activity against Trypanosoma cruzi and Leishmania infantum and high selectivities. Metallomics and interaction with BSA, apo-HTF and DNA were studied.

“…V V O-complexes were obtained in THF starting from [V IV O(acac) 2 ] and using equimolar ratio of reactants. Under the synthesis conditions, vanadium(IV) undergoes oxidation by molecular oxygen as previously reported for the homoleptic V V O-hydroxyquinoline derivatives [ 42 , 43 ].…”

“…Four bioactive oxidovanadium(V) heteroleptic compounds including 8HQ were developed and their biological behavior on T. cruzi was studied in detail [ 41 ]. More recently, two homoleptic series [V IV O(L-H) 2 ] and [V V O(OCH 3 )(L-H) 2 ], with L = 8-hydroxyquinoline derivatives, were studied in depth to shed light into the significance of the presence of the tridentate co-ligand on the biological behavior of the vanadium compounds [ 42 ]. Globally, results demonstrated the importance of the presence and nature of the tridentate coligand.…”

“…Besides the ability to cleave/damage DNA, the binding interactions of vanadium complexes to DNA have also been studied by different techniques; for example electronic absorption [56,[59][60][61][62], circular dichroism [60][61][62], and fluorescence emission have been used frequently [42,[60][61][62]65]; other techniques such as atomic force microscopy [66][67][68][69], viscosity [56,[59][60][61]69] and have also been applied. 51 V NMR, [53,54] capillary electrophoresis and Fourier transform infrared difference spectroscopy [70] has also been used, but much less frequently, as well as DNA melting, applied to evaluate e.g., binding and/or DNA crosslink formation by the complexes [56,59,60,71].…”

“…Vanadium is among most significant environmental contaminants; being a hapten, it exerts adverse impacts on the cardiovascular, respiratory, and reproductive systems and produces neurotoxic and immune-toxic effects [3]. But at the same time, the latest achievements in medicine such as creation of vanadium-containing orthodontic / orthopedic implants and development of anti-parasitic, antiviral, anti-bacterial, antithrombotic, anti-hypertensive, hypolipidemic, spermicidal, anti-tuberculosis, antitumor, and anti-diabetic medications make it necessary to resolve disputable issues related to influence exerted by vanadium on the immune system [4][5][6][7][8][9]. Adverse consequences caused by effects produced by chemical environmental factors on human health can become apparent not only via immunedependent diseases occurrence but also fail-ure in body adaptation mechanisms.…”
