“…2). These findings were consistent with previous studies of VCP expression in colorectal carcinoma, prostate cancer, oropharyngeal cancer, and lymphoma in which high expression of VCP was correlated with a lower 5-year survival rate [9,11,31,32]. In addition, subgroup analysis illustrated that VCP-positive breast cancer patients had worse OS than VCP-negative patients in both TNM stage I/II and III (Fig.…”

“…In human papillomavirus (HPV)‐negative oropharyngeal squamous cell carcinoma patients treated with surgery, VCP expression evaluated by immunohistochemistry indicated that this protein could be used as a prognostic marker, and VCP intensity was apparently uncorrelated with HPV status . Zhu et al recently investigated the correlation between VCP expression and primary orbital mucosa‐associated lymphoid tissue in lymphoma patients treated by curative resection; the results showed that high expression of VCP was significantly correlated with recurrence, enlarged tumor size, and reduced survival . The novelty of our analysis is that elevated expression of VCP was indicative of unfavorable survival in ESCC patients who received radiotherapy.…”

“…With respect to the high protein synthesis burden and possible aneuploidy in many kinds of tumor cells, the ERAD pathway has a central role in regulating cancer cell proliferation . Over the past few years, high expression of VCP has been described and found to correlate with disease outcomes in various cancer patients who received surgical resection . Recently, several VCP inhibitors have been developed .…”

“…VCP inhibitors perturb intracellular amino acid levels, activated eukaryotic translation initiation factor 2α kinase 4 (EIF2AK4), and enhance cellular dependence on amino acid supplies, consistent with a failure of amino acid homeostasis [65]. Thus, depletion of VCP triggers cancer cell death in part through inadequate control of protein synthesis and amino acid metabolism, and allows it to have implications for the development of anti-cancer therapies [65, 66]. OPG's interaction with VCP in the context of invasive and inflammatory breast cancer opens up many interesting avenues to research the therapeutic implications.…”
