Expression levels of 39 Cyp mRNAs in the mouse brain and neuroblastoma cell lines, C-1300N18 and NB2a – strong expression of Cyp1b1

Abstract: -An analysis of mRNA levels of 39 Cyp enzymes in the mouse brain and neuroblastoma cell lines, C-1300N18 and NB2a was performed using a real-time reverse transcriptase-polymerase chain reaction. Relative expression levels were quantified by normalized β-actin levels, and compared to the mRNA expression level of the cannabinoid receptor (CB1R), which is abundantly expressed in the mouse brain. Mean 2^-ddCts of CB1R in mouse brain, C-1300N18, and NB2a cells were 1.043, 1.003, and 1.005, respectively. Among Cyp m… Show more

“…More so, Hacl1 was not detected, which is in agreement with published data that shows Hacl1 mRNA to be virtually undetectable in the mouse brain [34]. None of the Cyp4a proteins detected and elevated in the liver where detected in the brain, which is also consistent with mRNA expression studies showing mouse brain Cyp4a10, Cyp4a12, and Cyp4a14 relative expression to be very low [35]. Cerebronic acid (2OH-C24:0) is a major 2-hydroxylated fatty acid component of cerebrosides and sulfatides in the brain and nervous tissues that undergoes peroxisomal α-oxidation.…”

“…Using targeted P450 proteomics, we identified a number of P450s in mouse, cynomolgus macaque, and human brain microsomes. Several studies have shown the presence of P450s, either at the mRNA or protein level, in murine brain (Stapleton et al, 1995;Choudhary et al, 2005;Renaud et al, 2011;Hersman and Bumpus, 2014;Stamou et al, 2014;Yamaori et al, 2017). Our identification of Cyp1a1, Cyp2c39, Cyp2d10, Cyp2d26, Cyp4a10, and Cyp7b1 coincides with these studies (Supp.…”

“…In contrast, CYP2E1 appeared to partially localize in the ER, as it has been reported in other cell types and tissues [ 3 , 53 , 83 ]. Recently, in other neuroblastoma cell lines, this isoform presented very low expression levels [ 84 ]. With the difficulty of the quantitative and qualitative analysis of the induction, this could explain why the fluorescence in some tested conditions did not reach the threshold required for the calculation of the R coefficient in our experiments.…”
