Expression of 3β-hydroxysteroid dehydrogenase in ovarian and uterine tissue during diestrus and open cervix cystic endometrial hyperplasia-pyometra in the bitch

Abstract: 24The purpose of this study was to compare the expression of 3β-hydroxystreroid 25 dehydrogenase (3β-HSD) in the uterus and ovary of healthy dogs and those with cystic 26 endometrial hyperplasia/pyometra complex (CEH-pyometra). Eighteen female dogs were 27 included in the study. Eleven bitches with open cervix CEH-pyometra were included in the 28 CEH-pyometra group and 7 diestrus bitches in the control group. For immunostaining a rabbit 29 polyclonal one raised against recombinant human type 2 (adrenal/gonadal… Show more

“…A negative effect by progesterone on the maturation of antigen‐presenting dendritic cells could also contribute to a diminished immune defence (Wijewardana et al., ). Increased expression of 3β‐hydroxysteroid dehydrogenase in the uterine endometrial tissue was recently shown in bitches with cystic endometrial hyperplasia–pyometra complex (Gultiken et al., ). These findings indicated that a local progesterone synthesis could be involved in the pathogenesis and promote the development of pyometra although circulating hormone levels are within the normal range.…”

“…Additionally, the pyometra‐promoting effects of exogenous progesterone may be potentiated if oestrogen is first administered and progesterone thereafter (Teunissen, ). The role of circulating hormones for the development of the disease has been debated, because most often concentrations are found to be comparable to those of healthy bitches at similar stages of the oestrous cycle (Gultiken, Yarim, Yarim, Gacar, & Mason, ). The effect may vary depending on different expression of hormone receptors (De Bosschere, Ducatelle, & Tshamala, ).…”

“…Wijewardana et al (2015) described negative correlation between progesterone on maturation of antigen presenting dendritic cells which may lead to reduction of cell mediated immunity (CMI). Gultiken et al (2016) reported increased expression of 3β-hydroxysteroid dehydrogenase on endometrial tissue in bitches suffering from pyometra which further indicates influence of local production of progesterone on disease occurrence even at its normal range. Therefore reduced CMI due to progesterone dominance in luteal phase (Sugiura et al, 2004), along with suppressed local immunity is favorable uterine environment for pathogens leading to their enhanced growth and colonization.…”
