Expression of betaine transporter mRNA: Its unique localization and rapid regulation in rat kidney

Abstract: Betaine is a major compatible osmolyte in the renal medulla. It is taken up into cells via the betaine gamma-amino-n-butyric acid transporter (BGT-1). We investigated the localization of BGT-1 mRNA and its acute regulation by NaCl and furosemide administration. In situ hybridization revealed that BGT-1 mRNA is predominantly present in the outer medulla and papilla. Less intense signals were seen in the inner medulla and no signals were found in the cortex. Microscopic examination suggested that intense signals… Show more

“…Myo-inositol is taken up by SMIT1, while sorbitol is synthesized from glucose by aldose reductase. Both SMIT1 (40) and aldose reductase (40,56) are concentrated in the medulla and have been shown to protect cell cultures derived from the inner medulla (2,56). However, reduction of sorbitol levels by inhibition of aldose reductase does not appear to cause serious kidney damage (60).…”

“…TonEBP stimulates genes coding for transporters and an enzyme that catalyzes cellular accumulation of organic osmolytes via concentrative uptake across plasma membrane and synthesis, respectively: the sodium/myoinositol cotransporter (SMIT) (40), the sodium/chloride/betaine cotransporter (BGT1) (28), and aldose reductase (AR) (15). The abundance of TonEBP (3) and mRNA of SMIT, BGT1, and AR (27) is much higher in the hypertonic renal medulla compared to the renal cortex or other tissues.…”

“…Renal BGT1 was cloned from Madin-Darby canine kidney (MDCK) cells (52) and is expressed in many mammalian tissues, including the brain (5,30). In the kidney, BGT1 mRNA is most abundant in the medullary thick ascending limb and the inner medullary collecting duct (31). The predicted structure of BGT1 contains 12 membrane-spanning ␣-helices with both NH 2 and COOH termini facing the cytoplasm.…”
