Expression of endogenous retroviruses, ERV3 and λ4–1, in synovial tissues from patients with rheumatoid arthritis

Abstract: SUMMARY We addressed the question of whether or not expression of human endogenous retroviruses (ERV). ERV3 and λ4–1, is related to the pathogenesis of rheumatoid arthritis (RA). In genomic Southern hybridization, there were no significant differences between RA patients and healthy volunteers with regard to frequencies of restriction fragment length polymorphism (RFLP) patterns, for either ERV3 or λ4–1. By Northern blot analysis using fresh synovial tissues, cultured synovial cells, and peripheral blood monon… Show more

“…Antibody cross-reactivity between exogenous retroviruses and ERV3 peptides have been described ( Katsumata et al, 1999 ) and ERV3 is up-regulated by cytokines in endothelial cells ( Sasaki et al, 2009 ). Indeed, ERV3 has been suggested as an auto-antigen involved in different immune-pathologies ( Takeuchi et al, 1995 ; Li et al, 1996 ; de Parseval et al, 1999 ; Blank et al, 2009 ; Nelson et al, 2010 , 2014 ; Kowalczyk et al, 2012 ). Expression of ERV3 was found to be up-regulated in blood cells but down-regulated in skin biopsies from patients with morphea ( Li et al, 1996 ).…”

“…Some CrERVs are transcriptionally active in mule deer [12]. Under normal physiological conditions, it is presumed that most ERVs are transcriptionally silenced by the host [9,16] and that failure to do so is associated with disease [17][18][19][20][21]. It is, therefore, extraordinary to have an outbred population in which transcriptionally active ERVs and insertional polymorphism are the norm.…”

“…The etiology is unknown, although possible causes include viral triggers 1 . The latter may include human endogenous retroviral (HERV) families: HERV-K, HERV-L, ERV-9, and ERV-3 2,3 . We have reported a novel reverse transcriptase polymerase chain reaction (RT-PCR) system to identify HERV-K10 and identified significantly high levels of messenger RNA in the peripheral blood (and synovial fluid) of patients with RA compared to osteoarthritis (OA) and healthy controls 4 .…”
