Expression of endothelial nitric oxide synthase gene in cultured porcine granulosa cells after FSH stimulation

Abstract: The present study was designed to investigate nitric oxide (NO) synthesis and the expression of endothelial NO synthase (eNOS) gene in cultured porcine granulosa cells. Granulosa cells prepared from small follicles (1-4 mm diameter) were cultured in plastic dishes coated with fibronectin in chemically defined medium, and matured after 48 h of stimulation with FSH. The concentrations of nitrite and nitrate remained relatively constant until 42 h of stimulation, after which they increased significantly up to two… Show more

“…Our results imply that eNOS-derived NO might also participate in the process but they might function differently. Since the expression of NOS might not be related to the synthesis of NO (Takesue et al, 2001), more direct evidence is needed to support the proposal.…”

“…The importance of NOS for meiotic maturation is confirmed by the expression of NOS isotypes in mouse and rat oocytes (eNOS, iNOSZackrisson et al, 1996;Jablonka-Shariff and Olson, 1997;Jablonka-Shariff et al, 1999;Nishikimi et al, 2001;Mitchell et al, 2004or nNOS mRNA -Abe et al, 1999 or in pig oocytes (eNOS -Hattori et al, 2000Takesue et al, 2001), and further confirmation of this comes from studies on mice with gene-knockout for eNOS. During maturation, oocytes of these mice exhibited blockage at the stage of metaphase I, or they exhibited various abnormalities (Jablonka-Shariff and Olson, 1998).…”

“…NOS isoforms also occur in the reproductive organs and oocytes of mammals (Tao et al, 1997;Jablonka-Shariff et al, 1999;Tesfaye et al, 2006). All three NOS isoforms likewise occur in pig oocytes during the growth period and meiotic maturation (Takesue et al, 2001 andChmelíková et al, 2009). The significance of NOS and NO for meiotic maturation was demonstrated in mammalian oocytes (Jablonka-Shariff and Olson, 1998;Schwarz et al, 2010).…”
