The primary cilium, a sensory apparatus, functions as both a chemical and mechanical sensor to receive environmental stimuli. The present study focused on the primary cilia in the epithelialmesenchymal interaction during tooth development. We examined the localization and direction of projection of primary cilia in the tooth germ and oral cavity of mice by immunohistochemical observation. Adenylyl cyclase 3 (ACIII)-immunolabeled cilia were visible in the inner/outer enamel epithelium of molars at the fetal stage and then conspicuously developed in the odontoblast layer postnatally. The primary cilia in ameloblasts and odontoblasts-shown by the double staining of acetylated tubulin and γ-tubulin-were regularly arranged from postnatal Day12, projecting apart from each other. The periodontal ligament possessed ACIII-positive cilia, which gathered on both sides of the dentin/cement and alveolar bone in postnatal days. In the oral cavity, numerous long primary cilia immunoreactive for ACIII were condensed at subepithelial stromal cells in the oral processes in fetuses, while postnatally a small number of short cilia were dispersed throughout the stroma of the oral cavity. These findings suggest that the primary cilia showing stage-and regionspecific morphology are involved in the epithelial-mesenchymal interaction during tooth development via mechano-and/or chemoreception for growth factors.

Rapid increase of knowledge in stem cell research, bioengineering technology and molecular basis of odontogenesis has finally lead us to the point where it is possible to develop approaches for treatment of tooth loss with bioengineered teeth, which one day might completely replace conventional prosthodontics and dental implants. By holding onto the premise that in order to bioengineer teeth, full understanding of how teeth develop is required, it must be acknowledged that there are certain features of odontogenesis which create obstacles in gaining that understanding. One such feature is the functional redundancy in genetic networks responsible for molecular control of odontogenesis. Abundant data imply that having functional redundancy of various elements in regulatory genetic networks is more than just a failsafe built into the odontogenic sequence in order to secure unhindered development of teeth. This phenomenon plays important roles in determination of tooth numbers and positioning, and is increasingly recognized as important for enabling sufficient plasticity of regulatory genetic networks through which the appearance of tooth-type specific and species-specific diversity of mammalian tooth morphology can be explained. Unfortunately, most of what we know about molecular basis of odontogenesis, comes from studies of mouse molars. Novel insights from developmental biology, stem cell research, and bioengineering technology are still needed, if we want to make de novo tooth bioengineering a clinically viable method for treatment of tooth loss.

Primary cilium is a solitary organelle that emanates from the surface of most postmitotic mammalian cells and serves as a sensory organelle, transmitting the mechanical and chemical cues to the cell. Primary cilia are key coordinators of various signaling pathways during development and maintenance of tissue homeostasis. The emerging evidence implicates primary cilia function in tooth development. Primary cilia are located in the dental epithelium and mesenchyme at early stages of tooth development and later during cell differentiation and production of hard tissues. The cilia are present when interactions between both the epithelium and mesenchyme are required for normal morphogenesis. As the primary cilium coordinates several signaling pathways essential for odontogenesis, ciliary defects can interrupt the latter process. Genetic or experimental alterations of cilia function lead to various developmental defects, including supernumerary or missing teeth, enamel and dentin hypoplasia, or teeth crowding. Moreover, dental phenotypes are observed in ciliopathies, including Bardet-Biedl syndrome, Ellis-van Creveld syndrome, Weyers acrofacial dysostosis, cranioectodermal dysplasia, and oral-facial-digital syndrome, altogether demonstrating that primary cilia play a critical role in regulation of both the early odontogenesis and later differentiation of hard tissue-producing cells. Here, we summarize the current evidence for the localization of primary cilia in dental tissues and the impact of disrupted cilia signaling on tooth development in ciliopathies.
