Expression of microRNA-150 and its Target Gene IGF2BP1 in Human Osteosarcoma and their Clinical Implications

Abstract: Previous study revealed that microRNA (miR)-150 might function as a tumor suppressor in osteosarcoma partially by targeting Insulin-Like Growth Factor 2 mRNA-Binding Protein 1 (IGF2BP1). The aim of this study was to investigate the clinical significance of miR-150-IGF2BP1 axis in human osteosarcoma which remains unclear. At first, expression levels of miR-150, and IGF2BP1 mRNA and protein in 20 osteosarcoma and matched adjacent noncancerous tissues were respectively detected by quantitative real-time PCR and w… Show more

“…We built regulatory networks with identified potential miRNA/gene and DMP/gene regulatory pairs, in which some pairs had been reported to exert positive effect on cancer pathogenesis and progression. For example, hsa-miR-150/ IGF2BP1 regulatory pair was reported to be a novel potential therapeutic target for osteosarcoma treatment ( 65 ), and IGF2BP1 was identified as a novel target gene of hsa-miR-98-5p in hepatocellular carcinoma ( 66 ). Similarly, the DNA demethylation in the promoter region of IGF2BP3 could influence the progression of G-CIMP gliomas ( 67 ), and cg07166550/ ALKBH5 could be used as prognostic biomarkers in prostate cancer ( 68 ).…”

“…In osteosarcoma, the expression of miR-10b and miR-146a has been related to tumor recurrence [62,63], and elevated levels of miR-21, miR-92a and miR-125b or low levels of miR-26a and miR-382 have been associated with poor prognosis [64,65,66]. Wang et al, have associated low levels of miR-150 with high tumor OS grade and poor response to chemotherapy [67].…”

“…First, we obtained 329 differentially expressed RBPs. Among them, CDC5L (40) and IGF2BP1 (41) have been indicated to be biomarkers of osteosarcoma and affect the prognosis of osteosarcoma. To elucidate the underlying biomolecular mechanism of differentially expressed RBPs, we used GO and KEGG enrichment analyses.…”
