Expression of P2X7 Receptor IncreasesIn VivoTumor Growth

Abstract: The P2X7 receptor is an ATP-gated ion channel known for its cytotoxic activity. However, recent evidence suggests a role for P2X7 in cell proliferation. Here, we found that P2X7 exhibits significant growth-promoting effects in vivo. Human embryonic kidney cells expressing P2X7 exhibited a more tumorigenic and anaplastic phenotype than control cells in vivo, and the growth rate and size of these tumors were significantly reduced by intratumoral injection of the P2X7 inhibitor-oxidized ATP. The accelerated growt… Show more

“…These conclusions were supported by experiments with siRNA. Similar effect of P2X7R on cell survival and effects of the allosteric inhibition of P2X7R were also shown on mouse pancreatic stellate cells,25 ovarian carcinoma cells38 and in vivo on melanoma cancer model 20…”

“…This validates that the P2X7R allosteric inhibitor AZ10606120 has anti‐proliferative effects both in vitro and in vivo . This inhibitor is not yet widely tested in cancer, but two studies show that similar anti‐proliferative effect of the inhibitor was detected in subcutaneous cancer induced by HEK293‐hP2X7 or B16 melanoma cells 1, 20…”

“…Tumors induced by the implant of HEK-293 cells heterologously expressing P2X7 receptors in immunodeficient mice showed elevated growth rate, an anaplastic phenotype, and high production of VEGF a well-known EMT inductor [67]. In cell lines derived from neuroblastoma or ovarian carcinoma, P2X7 receptor activity is correlated with high tumoral growth and metastatic induction, effects that involve PI3K/ AKT pathway activity [67][68][69][70]. Moreover, in pancreatic ductal adenocarcinoma cell lines, P2X7 receptor activity regulates migration and invasion, responses associated with EMT induction [71].…”

“…P2 receptors are also expressed in many types of cancer [9]. Among these molecules, P2X7 receptor shows increased expression in many types of cancer cells [10][11][12][13][14]. Furthermore, activation of P2X7 receptor has been implicated in tumor growth, as well as tumor angiogenesis and metastasis of breast cancer [6, 10-12, 15, 16].…”
