Expression of plant genes during the development of pea root nodules

Abstract: The expression of plant genes involved in the pea‐Rhizobium symbiosis was studied by analysing mRNA from root nodules. The RNA was translated in vitro and the translation products were separated by two‐dimensional gel electrophoresis. The results show differential expression of nodulin genes during root nodule development. One gene encoding N‐40′ is expressed at a significant level 5 days before the leghemoglobin genes. Most other nodulin genes are expressed more or less concomitantly with the leghemoglobin ge… Show more

“…The possibility to describe root nodule development not only by cytological criteria but also in molecular terms facilitates analysis of wild-type and mutant root nodule development. It has been recognized that nodulin genes are expressed at different time stages of nodule development (Govers et al, 1985;Gloudemans et al, 1987). This has led to the division between early and late nodulins.…”

“…Inoculated roots were harvested 4 and 8 days after inoculation; nodules were harvested 15 days after inoculation. Total RNA was extracted from plant tissues as described previously [ 14]. Fusarium oxysporum mycelium was inoculated in Czapek-dox medium and grown for 2 days at 30 ° C. Pea plants were inoculated with this suspension 3 days after sowing.…”

“…Nodulespecific host proteins are refered to as nodulins. Most of the nodulins described to date are induced at approximately the same time as leghemoglobin (9,10). However, several nodulins are induced before leghemoglobin in soybean and pea (8).…”

“…Translatable mRNA for nodulin N-75 is first detectable when soybean nodules are emerging through the epidermis, at 7 d after inoculation. Nodulin N-40' can be detected in pea roots 8 d after inoculation, 4 to 5 d before the onset of nitrogen fixation (10).…”
