Expression of proteins associated with adipocyte lipolysis was significantly changed in the adipose tissues of the obese spontaneously hypertensive/NDmcr-cp rat

Abstract: BackgroundThe etiology of the metabolic syndrome is complex, and is determined by the interplay of both genetic and environmental factors. The present study was designed to identify genes and proteins in the adipose tissues with altered expression in the spontaneously hypertensive/NIH –corpulent rat, SHR/NDmcr-cp (CP) and to find possible molecular targets associated with the pathogenesis or progression of obesity related to the metabolic syndrome.MethodsWe extracted RNAs and proteins from the epididymal adipo… Show more

“…In order for the intestine to absorb TAG, TAG must first be hydrolyzed trough a three-step process catalyzed by adipose triglyceride lipase, hormonesensitive lipase, and monoacylglycerol lipase (MGLL) 11 12 . In particular, recent studies showed that MGLL is located in villi, enterocytes and adipocytes and is involved in the final step of TAG hydrolysis, specifically, hydrolyzing monoglycerides to free glycerol and fatty acids 13 14 15 .…”

“…Nevertheless, at the mRNA levels an upregulation of hepatic transporter in biochips over the time of culture (PGP and OATP) was found. In parallel, an upregulation of PPAR alpha mRNA levels was found, which was reported to correlate with the downregulation of CES3 (carboxy esterases) . These last results can be related to the weak DCFDA expression in biochips because multidrug resistance transporters (PGP, OATP) would excrete the DCFDA and the CES activity would be too weak to produce fluorescence.…”

“…The selected spots were excised manually from Coomassie Brilliant Blue (CBB) R-350 (PhastGel Blue R, GE Healthcare)-stained preparative 2-DE gels with gel spot cutter. Then, in-gel digestion of protein samples was performed using the protocol described previously [ 17 ]. The peptide mixtures were analyzed by a matrix-assisted laser desorption ionization time-of-flight tandem mass spectrometry (MALDI-TOF/TOF MS; 4800 Plus MALDI TOF/TOF ™ Analyzer; Life Technologies, Carlsbad, CA) operating in positive-ion reflector mode.…”
