Expression of soybean-embryo lipoxygenase 2 in transgenic tobacco tissue

Abstract: To assess the role of lipoxygenase (LOX; EC 1.13.11.12) in plants, we increased the expression of LOX in the tissues of Nicotiana tabacum L. cv. 'KY 14' by over-expression of the LOX2 gene from the soybean (Glycine max (L.) Merrill) embryo. The LOX2 cDNA was manipulated by replacing its 5'-untranslated sequence with the translational enhancer of the alfalfa mosaic virus (AMV), and subcloned into a plant expression vector, 3' to a duplicated cauliflower mosaic virus 35S promoter. The AMV-LOX2 construct was tran… Show more

“…The availability of sufficient amounts of substrate seems to play a crucial role in the activation of the LOX pathway as tobacco plants overexpressing different LOX genes show no or only moderately elevated levels of LOX-derived metabolites (50,51). Because linoleic acid-derived oxylipins were the major derivatives detected in elicitor-treated potato cells, the linoleic acid pool was analyzed in two different ways: (i) by studying mRNA accumulation of ⌬ 12 -desaturases, which catalyze the formation of linoleic acid from oleic acid at the sn2 position of PC within the endoplasmic reticulum, and (ii) by measuring activity of PLA 2 , the major enzyme responsible for release of these PUFAs into the cytosol.…”

“…Hexenal, hexenol reduced to 1.5% of wild-type [329] Nicotiana tabacum LOX2 of soybean was overexpressed in tobacco under CaMV35S promoter. There was a 50-529% increase in C6 volatile aldehyde production [330] Solanum tuberosum The potato plastidic LOX H1 was silenced by antisense methodology. Volatile C6 aldehydes were reduced.…”
