Expression of the Androgen Receptor and its Correlation with Molecular Subtypes in 980 Chinese Breast Cancer Patients

Abstract: BackgroundRecent studies have shown that androgen displays an inhibitory effect on breast cancer cell lines that express androgen receptor (AR) but not estrogen receptor (ER) and progesterone receptor (PR). We have previously reported that approximately 1/3 of ER negative high grade invasive ductal carcinomas express AR. Thus, AR can serve as a potential therapeutic target for this group of patients.AimHere we investigated AR expression patterns in 980 consecutive breast carcinomas.ResultsWe found that (1) AR … Show more

“…In the present study, TNBC expressing AR had lower proliferation rate than AR-negative tumors (p<0.05). This finding is consistent with the results of recent studies using a 30% cutoff point 6 or mean Ki-67 values 24 . In our study, significance was found in the two methods of analysis; however, a higher cutoff point (50%) was used because 70% of TNBC cases showed Ki-67 labeling indices >50%.…”

“…The evaluation of histological grade showed no association with AR expression, a finding consistent with previous reports 6,24 . The attempt to subclassify AR expression into strong and weak showed no statistical significance in relation to Ki-67 or histological grade, and a possible explanation for this may be the extremely small number of patients with TNBC expressing AR positivity (n=11).…”

“…63 Although the precise role of AR in breast carcinogenesis is not clear, the possibility of it serving as a therapeutic target, especially in TNBC, is of significant interest. 35 Clinically, AR positivity is associated with better prognosis and, more recently, 21 associated with a lower recurrence score using the 21-gene test. 64 p53 By controlling the cell cycle and inducing apoptosis when cell damage is beyond repair, p53 is a vital regulator of genomic stability.…”

“…31,33,34 AR is expressed in up to 90% of breast cancers and up 50% of TNBCs. 35 AR expression is associated with better overall survival and disease-free survival, irrespective of coexpression of ER. 21,36,37 The positivity of p16, a tumor suppressor, is associated with better clinical outcome in some studies 38,39 but worse prognosis in others.…”
