Expression of the Ciona intestinalis Alternative Oxidase (AOX) in Drosophila Complements Defects in Mitochondrial Oxidative Phosphorylation

Abstract: Defects in mitochondrial OXPHOS are associated with diverse and mostly intractable human disorders. The single-subunit alternative oxidase (AOX) found in many eukaryotes, but not in arthropods or vertebrates, offers a potential bypass of the OXPHOS cytochrome chain under conditions of pathological OXPHOS inhibition. We have engineered Ciona intestinalis AOX for conditional expression in Drosophila melanogaster. Ubiquitous AOX expression produced no detrimental phenotype in wild-type flies. However, mitochondri… Show more

“…Yeast NDI1, amplified from genomic DNA, was cloned into a Drosophila P-element vector under the control of a GAL4-dependent promoter, flanked by insulator elements. Following microinjection (VANEDIS Drosophila Injection Service), transgenic progeny were established as independent lines NDI1 A46 and NDI1 B20 , with genomic insertion sites (on chromosomes 3 and 2, respectively) determined by inverse PCR (13). NDI1 A46 ∕3 and NDI1 B20 ∕2 denote these transgenes in combination with the corresponding wild-type chromosomes.…”
