Expression of the CXCL12/SDF-1 Chemokine Receptor CXCR7 in Human Brain Tumours

Abstract: Purpose: Receptor 7 (CXCR7) has recently been characterized as a novel receptor for CXCL12/SDF-1 (stromal cell derived factor-1). Given the demonstrated importance of CXCL12/SDF-1 in angiogenesis and tumour metastasis, we hypothesized that CXCR7 may also play a role in tumour pathogenesis. Located in the limited space of the intracranial cavity, any brain tumours can be inherently serious and life-threatening. However, the expression of CXCR7 in pituitary adenoma, neurilemmoma or hemangioblastoma remains to be… Show more

“…Barbieri et al have claimed that meningioma tissues express a number of receptors such as CXCR1, CXCR2, CXCR3, CXCR4, and CXCR5 among which CXCR4 had a mitogenic activity through ERK1/2 signalling pathway (Barbieri et al, 2006). CXCR7 mRNA was also detected in tumor samples from patients with neurilemmoma, pituitary adenoma, hemangioblastoma, and meningioma but the level of expression was significantly higher in neurilemmoma and meningioma compared to tissues from pituitary adenoma and hemangioblastoma (Tang et al, 2012).…”

“…ASCs are also CXCR4‐positive cells representing an additional target for SDF‐1 induced homing . Recently, a new SDF‐1 receptor named CXCR7 was identified on EPC, bone marrow‐derived MSCs, and in cancer cells of different origin, like ovarian, thyroid, and brain . In the latter, it plays an important role in tumor angiogenesis and metastatic dissemination.…”

“…In vivo and in vitro investigations have demonstrated that CXCR7 is widely distributed in neoplasms such as lung and cervical cancer, and brain tumour tissues; (3)(4)(5) furthermore, a higher level of expression of CXCR7 is linked to an early recurrence rate as well as a lower five-year disease-free survival rate in lung cancer patients. (3) The molecular mechanism of CXCR7 signalling acts through the enhancement of environmental tolerance and proliferation of tumour cells.…”
