To assess xenoestrogenicity, the 20 days-post-fertilization zebrafish (20dpfZF) life-stage was experimentally exposed to known model chemicals as well as to environmentally contaminated samples. From a selection of key responsive genes, molecular responses associated with the steroidogenic impact (brain aromatase, cyp19a1b and vitellogenin isoform 1, vtg1) were used to assess the endocrinic modulation by xenoestrogenic samples. This work showed diverse in vivo responses by the 20dpfZF, from pure chemicals (bisphenol A, BPA; 17 -ethinylestradiol, EE2; nonylphenol, NP; octylphenol, OP), and their mixture combinations, and to industrially contaminated sediment samples. Gene expression analysis of cyp19a1b and vtg1 in exposed 20dpfZF showed the estrogenic potencies of chemicals in the order: EE2 > OP > BPA > NP. Transcriptional up-regulation of vtg1 in early juvenile fish exposed to mixtures (BPA + NP + OP), relative to individual concentrations, suggest in minimum the additive in vivo effect of a mixture of very low sublethal doses of single xenoestrogens. Complementary expression of cyp19a1b and vtg1 in 20dpfZF, exposed to artificially-spiked sediment with EE2 established the estrogenresponsiveness of the model. In contrast, the lack of simple estrogenic or antiestrogenic trait of correspondence in biomarker transcription was evident from exposure to two types of industrially contaminated sediments, suggesting common non-linearity of in vivo response by the model. The results also highlight the applicability of 20dpfZF in the assessment of remediation and monitoring of historically polluted sites. This study showed that the 20dpfZF addressed the regulatory requirement of a multi-purpose and cost-effective ecotoxicological model capable of in vivo genomic assessment of endocrineactive substances in aquatic samples.

Abstract-The androgenic potential of a New Zealand pulp and paper mill effluent was measured by applying a combinatioll of in vitro and in vivo bioassays with mosquitofish (Gambusia a!finis) and goldfish (Carassius aurarus). The in vivo method assessed the rate of gonopodial development (masculinization) and alterations from normal reproductive behavior in adult female mosqllitofish exposed for 21 d to lIntreated 01' secondary-treated pulp mill effluent. A second in vivo mosquitofish exposure tested the effect of gl ass-fiber (type C) fi Itration of secondary-treated effIlient on rates of expression of the same endpoints. Extractable organics analyses of effluents and extracts thereof were conducted. Mosqllitofish demonstrated significant masculinization on exposure to either treated 01' untreated effluent; the frequency of gonopodial development was reduced with efiluent secondary-treatment. Male mating behavior was observed in the masculinized adult females. Glass-fiber (type F) filtration of the treated effluent eliminated the masclilinizing effect, suggesting that the bioactive compounds were associated with the suspended solids. The in vitra method measlired the binding of compounds within a treated thermomechanicallbleached kraft cffluent extract to androgen receptors contained in goldfish testis cytosol. Exposure to extracts of either the particulate (glass-fiber filtered) 01' the dissolved organic fraction of the efflllent produced significant binding (as indicated by the displacement of radiolabeled testosterone) to the androgen reeeptor in goldfish gonadal tissue. Thus, the dissolved organics extract of the treated effluent contained compounds androgenic to goldfish in vitro but not to mosquitofish in vivo. The combined in vitra and in vivo data suggest that the effluent in question could exert effects on the reproductive physiology of fishes through an androgenic mechanism. The androgenic compounds androstenedione and testosterone were not detected in the extracts used far the in vitro component of this study.

ABSTRACT. Phytosterols are potential endocrine-disrupting compounds. Quantification of phytosterols was carried out in sediments from four coastal zones and two rivers in south-central Chile. Sterol concentrations were determined by capillary gas chromatography-mass spectrometry and the sources of sedimentary organic matter were determined using sterol ratios and lipid biomarkers. Total sterol concentrations (0.03 to 10.4 µg g -1 ) were within the range reported for other marine ecosystems and the β-sitosterol concentration (0.01 to 2.01 µg g -1 ) was lower than previously reported for the upwelling system off Peru. Some coastal stations adjacent to the rivers had β-sitosterol of terrestrial origin. High concentrations of β-sitosterol were also found in sediments from more oceanic stations, supporting the notion that this sterol can also be produced by phytoplankton. No differences in the sterol concentration between the coastal zones were found. However, significant differences were found between almost all coastal zones and both rivers, and between rivers. At the station level and using different biomarkers of the source of organic matter, some areas were found to have a clear terrestrial influence; whereby it is assumed that the source of the phytosterols (especially β-sitosterol) would be vascular plants. The BioBío River and its mouth have a wide variety of sterols and lipids and high levels of cholesterol and epicholestanol, which is possibly related to the presence of domestic effluents derived from large cities. No clear spatial pattern emerge between the location of pulp mill industries and β-sitosterol sediment concentration, with the exception of one station located in the Gulf of Arauco. Los fitoesteroles son potenciales disruptores endocrinos. Se cuantificaron fitoesteroles en sedimentos de cuatro zonas costeras y dos ríos en el centro-sur de Chile. Se determinó la concentración de esteroles utilizando cromatografía de gas con espectrómetro de masa y las fuentes de materia orgánica sedimentaria se determinaron utilizando proporciones de esteroles y biomarcadores lipídicos. Las concentraciones de esteroles totales (0,03 a 10,4 µg g -1 ) se encuentran dentro del rango informado para otros ecosistemas marinos y la concentración de β-sitosterol (0,01 a 2,01 µg g -1 ) fue menor que la previamente informada para el sistema de surgencia de Perú. Algunas estaciones costeras adyacentes a los ríos presentaron β-sitosterol de origen terrestre.
