Expression of TLR3, 4, 7 and 9 in cutaneous malignant melanoma: relationship with clinicopathological characteristics and prognosis

Abstract: Toll-like receptors (TLRs) have achieved an extraordinary amount of interest in cancer research due to their role in tumor progression. The aim of this study was to investigate the expression and clinical relevance of TLR3, 4, 7 and 9 in cutaneous malignant melanoma (CMM). The expression levels of TLR3, 4, 7 and 9 were analyzed in tumors from 30 patients with CMM. The analysis was performed by immunohistochemistry, and the results were correlated with various clinicopathological findings and with relapse-free … Show more

“…These factors result in the resistance of tumor cells to immune cell attack and in evasion from immune surveillance (Huang et al 2005). Furthermore, high TLR4 expression was recently reported to be associated with shortened relapse-free sur- vival in melanoma (Eiro et al 2013) and with disease progression in colon cancer (Wang et al 2010). Our results show that TLR4 affects melanoma cell proliferation and motility, although LPS-induced proliferation was not inhibited robustly by TLR4 knockdown.…”

“…TLRs are a family of type I integral membrane glycoproteins, and based on its considerable homology in the cytoplasmic region, TLRs are also considered members of a larger superfamily that includes IL-1 receptors (IL-1Rs) (20). The human TLR family consists of 13 members, which are structurally characterized by the presence of a leucine-rich repeat domain in their extracellular region and a Toll/IL-1R domain in their intracellular region (21).…”

“…Through the adaptor molecule myeloid differentiation primary response protein 88 (MyD88), TLRs activate signaling pathways that are responsible for the induction of interferon (IFN) regulatory factor, activator protein-1 and nuclear factor kappa-light-chain-enhancer of activated B cells (20,24,25). The genes induced by the activation of TLRs produce several inflammatory cytokines, including tumor necrosis factor (TNF)-alpha, IFN-1, IL-6, IL-1 and granulocyte-colony stimulating factor (21). Different TLRs have been associated with the pathogenesis of several inflammatory and autoimmune skin diseases (26), and have gained interest in cancer research due to their role in tumor development and progression (18).…”

“…Stwierdzili, że wysoki poziom ekspresji TLR4 związany był ze znacznie krótszym czasem przeżycia pacjentów. Autorzy ci podkreślają, że wzrost ekspresji receptora TLR4 może być nowym niekorzystnym czynnikiem prognostycznym w czerniaku złośliwym [67]. Nieczerniakowe nowotwory skóry (ang.…”
