Expression of zebrafish ROR alpha gene in cerebellar‐like structures

Abstract: Mouse genetic studies have identified several genes involved in cerebellar development. The mouse mutants staggerer and lurcher are functionally deficient for the retinoid-related orphan receptor alpha (ROR alpha) and glutamate receptor delta2 (Grid2) genes, respectively, and they show similar functional and developmental abnormalities in the cerebellum. Here, we report the cloning and expression pattern of zebrafish ROR alpha orthologues rora1 and rora2, and compare their expression pattern with that of grid2… Show more

“…However, no isoform for these four genes were detected although RORa and RORg gene generate four and two isoforms in human, respectively [19]. In agreement with two RORa paralogues in grass carp, two RORa genes (also named RORa1 and RORa2) have been identified from zebrafish genome [14]. Although only one zebrafish RORg has been reported [13], another zebrafish RORg sequence can be found from NCBI database (GenBank ID: XM_685651).…”

“…Moreover, zebrafish RORa1 (also named RORA by Flores et al) is conserved in syntenic chromosomal relationships when compared with human RORa [13,14]. Similarly, a conserved syntenic relationship between zebrafish RORg1 and human RORg has been established.…”

“…About 100 genes were found upregulated or downregulated by at least twofold. Genes involved in the development of neurons and eyes, for instance, crx [30], rcv1 [31], neurod [32], vsx1 [33], rho [34], atoh2a [35] and rorab [36] were found downregulated (data not shown). Interestingly, the tumor suppressor p53 signaling pathway was found to be activated.…”

“…5). In zebrafish, the crest cells in the MON, Type I neurons in the optic tectum, and Purkinje cells express parvalbumin 7, glutamate receptor delta 2 (grid2), and retinoidrelated orphan receptor alpha 2 (rora2); the granule cells in these cerebellum-like structure express vglut1 (Mikami et al, 2004;Katsuyama et al, 2007;Bae et al, 2009) [Fig. 5(A-D)].…”
