“…PDE Enzyme Assays. The PDE activity was determined radiometrically as described previously (Wang et al, 1997) by SPA. The assay mixture contained 50 mM Tris, pH 7.5, 8.3 mM MgCl 2 , 1.7 mM EGTA, various concentrations of inhibitor, and an aliquot of the enzyme solution in a final volume of 100 l. After preincubation for 5 min at 30°C, the reaction was started by the addition of substrate (cAMP or cGMP).…”
