Expressions of adenosine A2A receptors in coronary arteries and peripheral blood mononuclear cells are correlated in coronary artery disease patients

Abstract: Expression of AR on PBMC correlated with those measured in coronary artery and aortic tissues in CAD patients, AR activity of PBMC matched that observed in aorta, and AR expression and activity in PBMC were found reduced as compared to controls. Measuring the expression level of AR on PBMC represents a good tool to address in situ expression in coronary tissues of CAD patients.

“…Although A 2A R are strongly expressed in the coronary system, it is difficult to investigate the adenosinergic pathway in the coronary artery. The fact that the expression and function of A 2A R in heart tissues30 and coronary arteries31 correlate with expression and function of A 2A R in PBMC provides a unique window to link the adenosinergic system to ischemia in the coronary arteries.…”

“…This may explain the lower level of A2A R in CS related to CHF compared with AHF. Finally, the lower A2A R expression in the AMI without CS group may result from the longstanding CAD condition and not from the acute myocardial ischemia (see references 28,38,39).…”

“…A 2A R expression measurement has been described previously (26,27). Peripheral blood mononuclear cells (PBMC) were used as the expression of A 2A R on PBMC correlates with those measured in coronary artery and aortic tissues (28). Blood samples were collected using Vacutainer CPT tubes (Beckton Dickinson, Franklin Lakes, NJ, USA).…”

“…Extracellular vesicles and PBMC were analysed by Western blotting as previously reported. [11][12][13][14][15] Briefly, freeze-dried EV corresponding to 100 µL of plasma and cell pellets corresponding to 0.25 × 10 6 PBMC were solubilized in 15 µL Laemmli sample buffer (BioRad) with 5% SDS, 5% 2-mercaptoethanol and a complete set of protease inhibitors (Roche), heated for 5 minutes at 95°C and loaded on 12% SDS-PAGE minigel (BioRad). Prestained molecular weight markers (BioRad) have always been loaded onto the gel.…”

“…The down‐regulation of A 2A R expression in CAD patients is related to the homocysteine (HCy) metabolism via its degradation product H 2 S . A 2A R expressed on peripheral blood mononuclear cells (PBMC) of CAD patients reflect coronary tissue expression showing the systemic nature of the adenosinergic signalling …”
